制备型色谱柱对黑豆皮花青素分离纯度的影响

【摘要】:以黑豆皮为原料,采用超声波辅助法提取3种黑豆皮花青素,利用高效液相色谱法对其进行定性定量分析,然后用两种不同型号的制备型色谱柱分离纯化含量最高的黑豆皮花青素,并鉴定其纯度。结果表明:3种黑豆皮花青素中的主要成分是矢车菊素-3-O-葡萄糖苷(C_3G),其中东北黑豆皮花青素中C_3G含量最高,为959.7 mg/100 g,其次是山西黑豆皮花青素。用两种色谱柱纯化东北黑豆皮花青素制备C_3G,色谱柱1 C18(77 mm×450 mm,10~100μm)分离的C_3G纯度为77.2%,色谱柱2 C18(19 mm×150 mm,5.0μm)分离的C_3G纯度为97.0%,色谱柱2分离的C_3G可以代替标品,适合实验室小量制备并做后续性质的研究。
[Abstract]:Three kinds of anthocyanins from black soybean peel were extracted by ultrasonic assisted method and analyzed qualitatively and quantitatively by high performance liquid chromatography (HPLC). Then the anthocyanins with the highest content were separated and purified by two different types of preparative chromatographic columns and the purity of anthocyanins was identified. The results showed that the main component of the three kinds of anthocyanins of black soybean peel was cornulin -3-O- glucoside C _ 3G _ (1), in which the content of C _ (3G) was the highest (959.7 mg/100 / g) in the anthocyanin of black soybean peel in Northeast China, followed by the anthocyanin of black bean peel in Shanxi Province. C3G was purified by two kinds of chromatographic columns. The purity of C3G was 77.2% for C _ (18) C _ (18) ~ (77) mm 脳 450 mm ~ (10 ~ (10) ~ (100 渭 m) for C ~ (3G), and 97.0 渭 m for C _ (3G) separation on column 2 C _ (18) ~ 19 mm 脳 150 mm ~ (5.0 渭 m). The C3G separated by column 2 could replace the standard product. Suitable for laboratory preparation and follow-up study.
