密蒙花中4个化合物含量的多波长高效液相色谱法测定

【摘要】:目的建立HPLC多波长法测定密蒙花中4个化合物(毛蕊花糖苷、蒙花苷、木犀草素、芹菜素)的含量,为密蒙花药材质量标准的建立提供参考。同时对不同产地密蒙花进行比较研究。方法采用高效液相色谱法,使用Waters Symmetry Columns(4.6mm×250mm,5 um)色谱柱,以甲醇(A)-0.1%磷酸水(B)为流动相,梯度洗脱(0-5min,5%A→40%A;5-15 min,40%A→50%A;15-20 min,50%A→63%A;20-35 min,63%A→100%A),流速0.8 ml·min~(-1),330nm(毛蕊花糖苷、蒙花苷),350nm(木犀草素),338nm(芹菜素),柱温25℃。结果在所建立色谱条件下各成分的专属性良好,无干扰峰;线性关系良好,均在0.9990以上;回收率在93.34%~104.69%,RSD均小于2%,回收率符合要求。测得6产地毛蕊花糖苷、蒙花苷、木犀草素、芹菜素的含量分别为0.3018~1.5269,2.0274~3.2220,0.0536~0.1214,0.3175~0.5590mg/g。结论经方法学验证,该法可在同一色谱条件下利用不同检测窗口实现多类组分及多个成分的同时测定。
[Abstract]:Objective to establish a multi-wavelength HPLC method for the determination of four compounds (Corlein, mongolin, luteolin, apigenin) in Mimonghua, and to provide a reference for the establishment of the quality standard of Mimonghua. At the same time, a comparative study on dense flowers from different habitats was carried out. Methods High performance liquid chromatography was used.
