HPLC法同时测定雪松不同部位5种酚酸类成分的含量

【摘要】:目的:建立同时测定雪松中没食子酸、原儿茶酸、儿茶素、咖啡酸和阿魏酸的含量测定方法,并用于雪松不同部位(松针、花序、嫩枝、木质部、树皮)该5种酚酸类成分的测定。方法:采用Eclipse Plus-C18(250 mm×4.6 mm,5μm)色谱柱,以乙腈(A)-0.1%磷酸水溶液(B)为流动相,梯度洗脱(0~6 min,7%A;6~70 min,7%A→20%A;70~80 min,20%A),流速1.0 m L·min~(-1),检测波长:280 nm,柱温:25℃。结果:没食子酸、原儿茶酸、儿茶素、咖啡酸和阿魏酸质量浓度分别在0.06~2.4、0.3~7.2、1.3~31.2、0.6~14.4和0.5~12μg·m L~(-1)范围内呈良好的线性关系,相关系数为0.999 6~0.999 9;平均加样回收率(n=6)分别为93.4%、102.2%、100.3%、99.6%和98.6%;精密度、重复性和稳定性良好,RSD均小于3%。雪松各部位中5种酚酸类化合物的含量存在明显差异,嫩枝中没食子酸、儿茶素和阿魏酸的含量最高,分别为0.134、2.345和0.838 mg·g~(-1),松针中原儿茶酸的含量最高为0.479 mg·g~(-1),木质中咖啡酸的含量最高1.113 mg·g~(-1),5种酚酸之和由高至低依次为嫩枝、木质、树皮、松针、花序。结论:经方法学验证,本法可用于雪松不同部位中5种酚酸类成分的含量测定。
