基于UPLC-Q-TOF-MS技术的芩百清肺浓缩丸血清药物化学初步研究

【摘要】:应用UPLC-Q-TOF-MS技术和血清药物化学理论分析检测芩百清肺浓缩丸的入血成分。灌胃给予大鼠芩百清肺浓缩丸溶液后,肝门静脉采血,血清经SPE处理,采用ACQUITY UPLC BEH C_(18)色谱柱(2.1 mm×100 mm,1.7μm),0.1%甲酸水(A)-0.1%甲酸乙腈(B)流动相系统进行梯度洗脱,流速0.3 m L·min~(-1),柱温35℃。通过对比分析体外样品、空白血清及含药血清,同时结合Peakview和Metabolitepilot软件鉴定和表征其血中移行成分,在给药后的大鼠血清中分析鉴定了28个血中移行成分,其中18个原型成分,10个代谢产物。应用UPLC-Q-TOF-MS技术和血清药物化学理论较为全面地阐明了芩百清肺浓缩丸在大鼠血清中的移行成分,为进一步研究其体内代谢过程以及药效物质基础提供了科学依据。
[Abstract]:The components of Qinbai Qingfei concentrated Pill were detected by UPLC-Q-TOF-MS technique and serum pharmacochemical theory. After the rats were given Qinbai Qingfei concentrated Pill solution by gavage, blood was collected from portal vein of liver and serum was treated with SPE. The gradient elution was carried out by ACQUITY UPLC BEH C _ (18) column (2.1 mm 脳 100 mm,1.7 渭 m) and 0.1% formic acid water (A) -0.1% acetonitrile (B) mobile phase system.
