苯并咪唑硫醚银配合物的合成及生物性质研究

【摘要】:由于金属银及其配合物具有良好的杀菌抗菌性质,近些年来引起了人们的兴趣与广泛关注。银离子的配位数从2到6,从而使它的几何构型既有平面线性构型,也有八面体构型。作为典型的杂环配体,苯并咪唑配体展现出了优良的光物理化学性质及良好的抗病毒性质。本论文选择苯并咪唑硫醚为配体,与不饱和烯酸银合成结构新颖的银配合物,并研究配合物结构与性质的关系。本文设计合成了四个苯并咪唑硫醚配体S1-S~4,与不同的烯酸银盐反应(甲基丙烯酸银,肉桂酸银,邻香豆酸银,对香豆酸银,水杨酸银,巴豆酸银),制备出七个不同的银配合物。用现代分析方法对其结构进行表征,七个不同的银配位化合物组成如下:[Ag(S~2)(p-coumarate)]·2CH_3OH(1)[Ag(S~3)(methacrylate)](2)[Ag(S~3)(cinnamate)](3)[Ag(S~3)_2](o-coumarate)·CH_3CN·2C_2H_5OH(4)[Ag(S~3)(crotonate)](5)[Ag(S~4)(crotonate)](6){[Ag_4(μ-4,4′-bipy)3(μ-salicylate)2(salicylate)H_2O]2?2C_2H_5OH}_n(7)X-射线单晶衍射表明配合物1、2、3、5、6结构相似,它们都是单核三配位的平面三角形结构,配合物4是单核四配位的扭曲四面体结构,配合物7是一个无机聚合物,并且是三核银簇化合物。尤其是在无机聚合物7中,每个聚合单位由中心对称的两部分组成,每个部分包含四个一价银离子,三个联吡啶配体,三个水杨酸阴离子和一个水分子,4,4′-联吡啶作为桥联配体与其他单元相连接,形成了无限延伸的一维链状聚合物。水杨酸阴离子有两种配位方式:一种是作为桥联配体与银(I)进行双齿配位,连接Ag1,Ag3,Ag_4;另外一种是与Ag2单齿配位,提供配位点。以Ag1为中心,周围几个原子形成了扭曲的四方锥体,四方锥的变形参数是τ5,用于形容五配位的金属体系变形系数。以Ag1为中心的四方锥参数τ5的数值是0.11;Ag2是三配位的,与周围原子构成了一个平面三角形;Ag3和Ag_4都是四配位的,与周围的氮氧原子形成四面体几何构型。应用紫外滴定,荧光滴定,粘度测试等实验方法,研究了配体、配合物与DNA之间的相互作用模式,测定了减色率,线性拟合常数Kb,荧光淬灭常数Ksv,实验数据表明,配体及配合物与DNA的作用模式是插入模式,配合物与DNA的结合能力要强于配体。使用Fetion法及NBT光照法研究了银配合物抗羟基自由基和抗超氧自由基的活性,用IC50值表征配合物清除自由基能力的大小。研究结果表明配合物1、4、5、7的IC50值比标准的VC和甘露醇低两个数量级,说明配合物1、4、5、7有更优异的清除羟基自由基的能力;而配合物1、7的IC50值比SOD标准模拟复合物小,说明配合物1、7可以作为SOD模拟酶清除超氧阴离子。
[Abstract]:Silver and its complexes have attracted much attention in recent years due to their good bactericidal and antibacterial properties. The coordination number of silver ions ranges from 2 to 6, which results in both plane linear configuration and octahedral configuration. As typical heterocyclic ligands, benzimidazole ligands exhibit excellent photophysicochemical and antiviral properties. In this paper, benzimidazole sulfides were selected as ligands to synthesize novel silver complexes with silver unsaturated enoate, and the relationship between the structure and properties of the complexes was studied. In this paper, four benzimidazolyl thioether ligands S _ (1-S) _ (4) were designed and synthesized.
