高效毛细管区带电泳法同时检测3个产地沙棘果粉中的活性物质
[Abstract]:Four active substances (rutin, kaempferol, isorhamnetin and quercetin) in Hippophae rhamnoides powder from three producing areas were separated and determined by high performance capillary zone electrophoresis (HPCE). The effects of different electrophoretic conditions on the separation efficiency were investigated, and the optimum electrophoretic conditions were obtained as follows: buffer solution 20 mmol/L Na_2B_4O_7-H_3BO_3 (mass concentration 1.5 mg/mL 尾-cyclodextrin), pH 9.55, p-cyclodextrin), pH 9.55, mass concentration 1.5 mg/mL), Detection wavelength 370 nm, separation voltage 25 kV, injection time 5 s. Under these conditions, four active substances, rutin, kaempferol, isorhamnetin and quercetin, were separated in 11 min. The linear ranges were 0.01 ~ 0.51, 0.05 ~ 0.93, 0.02 ~ 0.65, 0.03 ~ 0.81 mg/mL, respectively. The correlation coefficients were 0.997 脳 10 ~ (- 5) mg/mL (RSN=3), and the detection limits were 5.05 脳 10 ~ (- 5), 2.10 脳 10 ~ (- 5), 3.75 脳 10 ~ (- 5) and 1.31 脳 10 ~ (- 5) respectively. The intra-day and inter-day precision ranges are 0.08% and 4.72% respectively. The average recovery is between 95.51% and 104.66%. The relative standard deviation is less than 3.92% (n = 3). Under the optimized conditions, the mixture of four kinds of active substances was determined, and the extract of active substances from three producing areas of seabuckthorn fruit powder was detected by the same method. The results showed that the contents of four compounds in Hippophae rhamnoides from different producing areas were different. The highest total amount of four compounds in Hippophae rhamnoides was from Urumqi, Xinjiang, and the lowest was from Wuwei, Gansu Province. The quercetin content was the highest in Lv Liang, Shanxi, and the highest in kaempferol and isorhamnetin in Xinjiang Urumqi. The method is rapid, simple, accurate and reliable, and can be used for simultaneous determination of various active substances in seabuckthorn fruit powder.
