土大黄游离蒽醌类成分HPLC指纹图谱

【摘要】:目的:建立不同产地土大黄中游离蒽醌类成分HPLC指纹图谱的共有模式,评价31批不同产地土大黄药材的质量状况。方法:采用高效液相色谱法,Diamonsil C18色谱柱(4.6 mm×200 mm,5μm),以甲醇-0.2%甲酸溶液为流动相进行梯度洗脱,流速1.0 mL·min~(-1),柱温30℃,检测波长254 nm。采用"中药色谱指纹图谱相似度评价系统"(2004 A版)进行相似度评价,采用SPSS 18.0结合聚类分析(cluster analysis,CA)和主成分分析(principal component analysis,PCA)对31批土大黄药材进行质量评价。结果:标定共有指纹峰6个,并指认出大黄素(4号峰),大黄酚(5号峰)和大黄素甲醚(6号峰)3个成分。土大黄指纹图谱的精密度、稳定性、重复性中6个共有峰的相对保留时间与相对峰面积的RSD分别2.0%,5.0%。采用聚类分析法和主成分分析法将土大黄药材分为2类,其中有27批药材相似度在0.8以上,进而说明了31批土大黄药材之间存在相似性和差异性。结论:指纹图谱结合化学模式识别的构建有利于全面、准确地控制土大黄药材的内在质量。
