2-羟基-1,4-萘醌参与的多组分反应

【摘要】:萘醌类化合物是一类广泛存在于自然界中的小分子化合物,具有多种生物活性。其中2-羟基-1,4-萘醌结构在各种天然产物中是最常见的,并且与多种生物活性相关,包括酶抑制,抗真菌,抗细菌,抗癌,抗增殖,抗血小板,抗炎,抗过敏和抗疟疾活性等。基于2-羟基-1,4-萘醌的多种性能,我们以其为原料,使用一锅法将2-羟基-1,4-萘醌应用到多组分反应中,并且经过实验探究后,为合成3,4-二氢-2H-萘并[2,3-e][1,3]VA嗪-5,10-二酮衍生物、7-芳基苯并[c]吖啶-5,6-二酮衍生物、4H-苯并[g]色烯衍生物提供了新方法。水是最清洁的溶剂,也是最廉价的溶剂,在水介质中的反应,有利于降低反应成本,减少有机溶剂对环境造成的污染,这些优点使得水成为有机合成中溶剂的首选。我们以水做溶剂,室温条件下,用超声辐射2-羟基-1,4-萘醌,甲醛溶液,芳香胺,合成了一系列的3,4-二氢-2H-萘并[2,3-e][1,3]VA嗪-5,10-二酮衍生物。该反应的最大特点是以水做溶剂,不需要使用催化剂,采用超声辐射的方法,缩短了反应时间,提高了反应收率。近年来,金属有机框架化合物由于其在突光、磁体、气体选择性吸附、催化剂和光学性能上的潜在应用和其构成了多样的拓扑结构,得到人们的广泛重视。我们自己制备了金属有机骨架MOF-5材料,并以其为催化剂,乙醇为溶剂,在室温下催化了以醛、芳香胺、2-羟基-1,4-萘醌为原料的一锅法-三组分的缩合反应,开发了一种合成7-芳基苯并[c]吖啶-5,6-二酮衍生物新方法,且收率较高。在过去几年中,可见光促进的化学反应受到相当大的关注,并且成为有机化学中的热点研究课题。可见光是一种有吸引力,可持续,无毒,易于使用和普遍可用的能源,且光催化反应通常在常温常压下进行,较易控制,无二次污染,结合光催化和无溶剂反应的优势,我们开始了环境友好的战略,探索出了一条合成4H-苯并[g]色烯衍生物的新路线。通过2-羟基-1,4-萘醌,丙二腈/氰基乙酸乙酯,醛应用光催化,无溶剂的条件下使用Eosin Y做催化剂的一锅多组分反应。令我们惊喜的是,该方法不但提高了反应产率,缩短了反应时间,而且反应不需要使用溶剂,无有毒物质排出等优点均符合绿色化学的要求。
