超滤法提取银杏叶和绞股蓝双中药抗心血管疾病的有效成分

【摘要】:随着肥胖、不合理膳食、体力活动不足等心血管疾病危险因素的增加,心血管疾病发病率和死亡率在逐年递增,心血管疾病因存在发病率高,治愈率低,并发症多等特点,让其防治工作有了较大困难。传统医学和现代药理研究表明,与西药相比,活血化瘀作用的中药具有疗效好,活性强,副作用小等优点,在治疗心血管疾病方面具有独特优势。目前,单独的银杏叶和绞股蓝提取物制成胶囊、口服液、注射液、气雾剂等治疗心血管疾病药剂临床应用很多,银杏叶和绞股蓝的混合提取物和制剂少见报道。本文利用超滤法对所提取物分子量大小可控的特点,提取银杏叶和绞股蓝双中药中抗心血管疾病的有效成分,并与单独提取抗心血管疾病有效成分相比较,考察单味中药提取及双中药提取抗心血管疾病有效成份变化情况;试图得到比单中药更多的抗心血管疾病有效成份提取物,以充分发挥中药多成分多靶点协同作用的特点,为制备比单中药制剂治疗效果更好的抗心血管疾病药物提供理论基础。具体工作如下:1、以银杏叶和绞股蓝为研究对象,不同浓度乙醇水为提取液对银杏叶和绞股蓝进行单独粗提和混合粗提,然后抽滤得粗提液;2、用超滤设备对粗提液进行超滤提取,超滤液经减压浓缩、真空干燥得到所需产品;3、在超滤提取工艺研究中,采用单因素试验设计,考察了乙醇浓度、超滤压力、超滤温度和超滤时间对银杏叶和绞股蓝单独提取物和混合提取物中抗心血管疾病有效成分含量的影响,并确定主要影响因素;4、在此试验结果的基础上,通过正交试验设计,从单中药提取和双中药提取两个方面考察了银杏叶和绞股蓝超滤提取物抗心血管疾病有效成分含量的影响,并确定最佳工艺条件;5、采用分光光度法对提取物中的抗心血管疾病有效成份包括银杏黄酮、绞股蓝黄酮、绞股蓝皂苷进行含量测定,采用高效液相色谱法对银杏内酯进行含量测定,并进行比较分析。本研究结果显示:1、银杏叶、绞股蓝及双中药超滤提取抗心血管疾病有效成分最佳工艺条件均为乙醇浓度为50%,超滤压力为9.0 bar,超滤温度为30℃。2、在最佳超滤提取工艺条件下,单味银杏叶提取物黄酮含量为5.13%。单味绞股蓝提取物黄酮含量为4.52%,银杏叶和绞股蓝双中药提取物中黄酮含量为6.35%;单味银杏叶提取物总内酯含量为0.3424mg/g,银杏叶和绞股蓝双中药提取物中总内酯含量为0.3328mg/g;绞股蓝提取物皂苷含量为3.4%,银杏叶和绞股蓝双中药提取物中皂苷含量为3.52%。
[Abstract]:With the increase of risk factors of cardiovascular disease, such as obesity, unreasonable diet and insufficient physical activity, the incidence and mortality of cardiovascular disease are increasing year by year. The incidence of cardiovascular disease is high and the cure rate is low. Traditional medicine and modern pharmacological studies show that compared with western medicine, traditional Chinese medicine has better curative effect, stronger activity and less side effect than western medicine. At present, the single extract of Ginkgo biloba and gynostemma pentaphyllum is made into capsule, oral liquid, injection, aerosol and so on. Rare reports on the mixed extracts and preparations of Ginkgo biloba and gynostemma pentaphyllum. The effective components of Ginkgo biloba and gynostemma pentaphyllum were extracted from Ginkgo biloba and gynostemma pentaphyllum by ultrafiltration method. And compared with single extraction of effective components of cardiovascular disease, the changes of anti-cardiovascular effective components of single Chinese medicine extraction and double traditional Chinese medicine extraction were investigated. In order to give full play to the multi-component and multi-target synergistic action of traditional Chinese medicine, we try to obtain more effective extracts of anti-cardiovascular disease than single Chinese medicine. In order to provide the theoretical basis for the preparation of anti-cardiovascular diseases drugs which are better than the single Chinese medicine preparation, the specific work is as follows: 1, Ginkgo biloba leaves and gynostemma pentaphyllum as the research object. Ginkgo biloba leaves and gynostemma pentaphyllum were extracted separately and mixed with different concentrations of ethanol water. (2) the crude extract was extracted by ultrafiltration equipment, and the product was obtained by vacuum drying. 3. In the study of ultrafiltration extraction process, the concentration of ethanol and the pressure of ultrafiltration were investigated by single factor experimental design. The effects of ultrafiltration temperature and ultrafiltration time on the contents of effective components of anti-cardiovascular disease in single extract and mixed extract of Ginkgo biloba leaves and gynostemma pentaphyllum, and the main influencing factors were determined. 4. On the basis of the experimental results, the effects of extract of ginkgo biloba and gynostemma pentaphyllum on the content of effective components in cardiovascular disease were investigated from two aspects: single Chinese medicine extraction and double traditional Chinese medicine extraction by orthogonal design. And determine the best process conditions; 5. The contents of ginkgo flavonoids, gynostemma flavonoids and gynostemma saponins in the extracts were determined by spectrophotometry. The content of ginkgolide was determined by high performance liquid chromatography. The optimum conditions for extraction of effective components from cardiovascular disease by ultrafiltration of gynostemma pentaphyllum and traditional Chinese medicine were ethanol concentration of 50 kb, ultrafiltration pressure of 9.0 barand ultrafiltration temperature of 30 鈩,
