高效溶磷菌的筛

【摘要】:为了从根际土壤中筛选出具有较高溶磷能力的菌株及优化溶磷条件,通过种子萌发初步研究溶磷促生效应。通过稀释涂布法分离、筛选菌株,并进行ATB细菌鉴定仪及16S r DNA测序鉴定;采用单因子试验及正交试验优化菌株溶磷发酵条件,利用种子发芽指标测定菌株的促生能力。结果显示,筛选出菌株BD-1的溶磷能力最强,经ATB细菌鉴定仪及16S r DNA测序,该菌株被鉴定为路德维希肠杆菌。正交试验结果表明,在温度为30℃、摇床转速为180 r/min、接种量为2 m L、初始p H值为7.0的条件下培养7 d的溶磷效果最好,其溶磷量为181.73 g/L。种子萌发试验结果表明,该溶磷菌对作物种子萌发具有一定的促进作用。
[Abstract]:In order to screen the strains with high phosphorus solubilization ability from rhizosphere soil and optimize the phosphorus solubilizing conditions, the effect of phosphorus solubilization on growth was studied preliminarily through seed germination. The strains were isolated and screened by dilution coating method, and identified by ATB and 16s r DNA sequencing. The fermentation conditions were optimized by single factor test and orthogonal test, and the growth promoting ability of the strain was determined by seed germination index. The results showed that the strain BD-1 had the strongest phosphorus solubilizing ability. The strain was identified as Enterobacter Ludwig by ATB bacteriometer and 16s r DNA sequencing.
