1株高产IAA菌株的筛

【摘要】:从番茄根际采集的土壤中分离筛选得到12株产IAA的菌株,经复筛获得1株具有较好IAA生产能力的细菌,其发酵24 h产IAA水平达到48 mg/L以上,将其命名为HB-1。通过对菌株HB-1的16S r DNA序列分析对该菌进行鉴定,并将菌株HB-1进行液体发酵扩繁,将得到的发酵液冲施白菜,试验其促生作用。结果表明,该高产IAA菌株为枯草芽孢杆菌。盆栽试验结果表明,菌株HB-1对白菜生长具有明显的促生作用,施加HB-1发酵液处理的叶绿素含量、叶片数均高于对照(CK);HB-1发酵液处理的鲜质量、干质量与冲施清水的CK相比都增加,鲜质量增加1.54~17.98 g,增幅达1.16%~13.55%,差异达到显著水平,干质量增加0.26~1.32 g,增幅达到2.97%~14.92%。其中添加发酵液1.0 m L/盆的处理组效果较好,叶绿素含量比CK高5.02%,叶片数比CK高15.78%,鲜质量较空白处理增加17.98 g,增幅达13.55%,差异达极显著水平,干质量平均增加1.32 g,增幅达到14.92%。
[Abstract]:Twelve strains producing IAA were isolated from the soil collected from tomato rhizosphere, and one strain with good IAA production capacity was obtained by repeated screening. The level of IAA production reached more than 48 mg/L after 24 h fermentation, and it was named HB-1. The 16s r DNA sequence analysis of strain HB-1 was used to identify the strain, and the strain HB-1 was expanded by liquid fermentation. The results showed that the high IAA strain was Bacillus subtilis. The results of pot experiment showed that strain HB-1 had obvious promoting effect on the growth of Chinese cabbage. The chlorophyll content and leaf number of treated with HB-1 fermentation broth were higher than that of CK treated with (CK) HB-1 fermentation broth, and the dry weight was higher than that of CK treated with fresh water. The fresh weight increased by 1.54 ~ 17.98 g, the increase reached 1.16 ~ 13.55, the difference reached a significant level, the dry weight increased 0.26 ~ 1.32 g, and the increase reached 2.97% 14.92%. The effect of adding fermentation broth 1.0 mL / basin was better, chlorophyll content was 5.02% higher than CK, leaf number was 15.78% higher than CK, fresh weight increased 17.98 g, and the increase was 13.55%, the difference reached very significant level, dry weight increased 1.32 g on average, and the increase reached 14.92%.
