甜樱桃根际IAA产生菌的筛

【摘要】:为了给甜樱桃微生物肥料的研制和生产提供优良菌株资源,本文从马哈利樱桃根际土壤中分离获得52株细菌,通过保绿法和萝卜子叶增重法进行初筛,筛选具有促生作用的菌株,进一步进行IAA产生菌的筛选,并经形态观察、生理生化特征及16SrDNA序列分析明确分类地位,利用分光光度法测定产IAA的能力,并对最佳碳氮源、产素条件进行了筛选。结果表明,52株菌中B20产IAA能力最强,经鉴定为阿氏芽孢杆菌(Bacillus aryabhattai),其最佳产素的培养条件为:采用30%的装液量在温度为30℃、转速为150r·min-1的条件下培养44h,最佳碳、氮源分别为果糖和酵母粉。研究发现菌株B20具有IAA产生能力,具有作为功能微生物肥料研发的潜力,可进一步对其进行开发研究。
[Abstract]:In order to provide excellent strain resources for the development and production of sweet cherry microbial fertilizer, 52 strains of bacteria were isolated from the rhizosphere soil of Mahari cherry and screened by green preserving method and radish cotyledon weight gain method. Further screening of IAA producing strains was carried out, and the taxonomic status was determined by morphological observation, physiological and biochemical characteristics and 16SrDNA sequence analysis. The ability of producing IAA was determined by spectrophotometry, and the optimum carbon and nitrogen sources were analyzed. The production conditions were screened. The results showed that B20 had the strongest ability to produce IAA among 52 strains. The optimum conditions for producing IAA were as follows: the best culture conditions were as follows: 30% liquid loading at 30 鈩,
