N-乙酰半胱氨酸对脓毒症大鼠肠损伤的保护作用及对炎症因子表达的影响

【摘要】:为探讨N-乙酰半胱氨酸(NAC)对脓毒血症大鼠肠损伤的影响及对炎症因子表达的影响,选取清洁级SD大鼠30只,体重250~300g,随机分为假手术组(Sham组)、脓毒症组(SEP组)和N-乙酰半胱氨酸组(NAC组),每组10只;Sham组仅开腹,暴露盲肠,NAC组和SEP组行盲肠结扎穿孔术,再分别静脉给予150mg·kg~(-1)NAC和等量生理盐水;3组均于再灌注4h处死大鼠,观察小肠组织病理学结果;ELISA检测血中肠脂肪酸结合蛋白(I-FABP)和D-乳酸浓度;ELISA法和Western blot法检测血清和小肠中TNF-α、IL-6、IL-10浓度。结果表明:光镜下Sham组未出现病理损伤,SEP组损伤重,NAC组损伤减轻;与Sham组比较,SEP组I-FABP、D-乳酸显著升高(p0.05);与SEP组比较,NAC组I-FABP、D-乳酸显著降低(p0.05);与Sham组比较,SEP组血清中TNF-α、IL-6、IL-10显著升高(p0.05);与SEP组比较,NAC组TNF-α、IL-6显著降低(p0.05),IL-10显著升高(p0.05),小肠炎症相关因子变化趋势同血清检测。N-乙酰半胱氨酸对脓毒血症大鼠肠黏膜屏障有保护作用,其机制与抑制炎症反应有关。
[Abstract]:To investigate the effects of N-acetylcysteine (NACN) on intestinal injury and the expression of inflammatory cytokines in sepsis rats, 30 SD rats of clean grade were selected, weighing 250 ~ 300g. They were randomly divided into sham-sham group, septic group (SEP group) and N-acetylcysteine group (NAC group, n = 10 in each group). In the Sham group, the caecum ligation and perforation were performed in the exposed caecum group and the SEP group, and were given 150 mg 路kg~(-1)NAC and the same amount of normal saline respectively. All the rats in the 3 groups were killed at 4 h after reperfusion, and the histopathological results of small intestine were observed. The concentrations of serum fatty acid binding protein (I-FABP) and D-lactic acid were detected by ELISA. ELISA and Western blot were used to detect the concentration of IL-10 in serum and small intestine. The results showed that there was no pathological injury in Sham group under light microscope. The damage of SEP group was alleviated. Compared with Sham group, I-FABPnD-lactic acid in SEP group was significantly higher than that in Sham group (P 0.05). Compared with SEP group, I-FABP- D- lactic acid in NAC group was significantly lower than that in SEP group. Compared with the Sham group, the serum TNF- 伪 IL-6 + IL-10 in the Sham group was significantly higher than that in the Sham group (P 0.05). Compared with SEP group, TNF- 伪 IL-6 in NAC group decreased significantly (P 0.05) and IL-10 increased significantly (p 0.05). The change of inflammatory related factors in small intestine is related to the protective effect of serum. N-acetylcysteine on intestinal mucosal barrier in rats with sepsis, and its mechanism is related to the inhibition of inflammatory reaction.
