【摘要】:目的研究杀伤细胞免疫球蛋白样受体(KIR)与人类白细胞抗原(HLA)Ⅰ类在云南独龙族人群中的基因频率及配对分布。方法从云南独龙族无偿献血人群中选取157名健康无关志愿者,采用聚合酶链式反应-序列特异性引物扩增(PCR-SSP)进行KIR基因检测;采用聚合酶链式反应-直接测序分型法(PCR-SBT)进行HLA-A、B、C等位基因分型。KIR基因携带者频率和HLA-A,-B和-C等位基因频率以直接计数法计算。结果在所有检测的样本中,HLA-C1和HLA-C2频率分别为92.4%和7.6%。含有Bw4基序的HLA-A、-B基因频率分别为5.1%和6.4%。所有个体均含有抑制性KIR/HLA-C配受体对,含有1、2、3对KIR/HLA-C配受体对的频率分别为87.2%、8.3和4.5%。在KIR/HLA-Bw4配对中,KIR3DL1+HLA-Bw4配受体对频率为12.1%,KIR3DS1+HLA-Bw4配受体对频率为1.3%,2者均含有的频率为5.1%,2者均没有的频率为81.5%。结论云南独龙族人群HLA及KIR/HLA配受体对有独特的分布特征,抑制性的KIR-HLA配受体对占主导地位。
[Abstract]:Objective to study the relationship between killer cell immunoglobulin-like receptor (kir) and human leukocyte antigen (HLA). Methods 157 healthy unrelated volunteers were selected from volunteer blood donors of Dulong nationality in Yunnan Province. Polymerase chain reaction-sequence specific primer amplification (PCR-SSPs) was used to detect KIR gene. Polymerase chain reaction (PCR) -direct sequencing typing (PCR-SBT) was used to detect the frequency of allelic alleles. KIR and HLA-A. The frequencies of -B and -C alleles were calculated by direct counting. The results were found in all tested samples. The frequencies of HLA-C1 and HLA-C2 were 92.4% and 7.6, respectively. HLA-A with Bw4 motif was found. The frequencies of -B gene were 5.1% and 6.4, respectively. The frequencies of the pair of KIR/HLA-C receptor pairs were 87.2 and 4.5, respectively. The frequency of KIR3DL1 HLA-Bw4 receptor pair was 12.1and that of KIR3DS1 HLA-Bw4 pair was 5.1%. Conclusion the distribution of HLA and KIR/HLA receptor pairs in Dulong ethnic group is unique, and the inhibitory KIR-HLA receptor pair is dominant.
