【摘要】:背景:随着现代病理技术的不断创新及发展,在疾病诊断方面显著减少其误诊率,但特殊染色法依然是病理诊断的最主要手段。目的:比较不同的特殊染色方法显示人腰椎小关节结构的优缺点。方法:收集腰椎手术患者的L4/5小关节软骨标本,行苏木精-伊红染色、番红O染色、甲苯胺蓝染色、Masson染色、番红-固绿染色,观察软骨结构。结果与结论:苏木精-伊红染色、番红-固绿染色和甲苯胺蓝染色均可清晰观察到关节软骨各层结构。其中苏木精-伊红染色能较清楚显示软骨层、潮线及软骨下骨,软骨细胞核呈蓝紫色。番红-固绿染色清晰显示软骨4层结构:浅层(软骨表面),中间层(细胞圆形,着色较淡,排列规律),柱状细胞层(细胞较大,多核,排列整齐),显示软骨基质呈均匀浅红,软骨下骨呈绿色,软骨组织与骨组织分对比鲜明。甲苯胺蓝染色显示组织结构层欠清,胞核染色清晰,胞浆几乎不着色,基质呈淡蓝紫色。而番红O染色软骨层及软骨下骨层分界比较清晰,软骨层呈均匀深红色,细胞染色较淡,与软骨基质分界不明显。Masson染色对胶原纤维较为清晰,但只能大致显示软骨与软骨下骨结构。结果说明番红-固绿染色显示软骨各层和软骨下骨结构最佳;苏木精-伊红染色观察软骨细胞形态变化较其他方法清楚,Masson染色最次。
[Abstract]:Background: with the continuous innovation and development of modern pathological technology, the misdiagnosis rate of disease diagnosis has been significantly reduced. But the special staining method is still the most important method for pathological diagnosis. Objective: to compare the advantages and disadvantages of different special staining methods in the structure of human lumbar facet joints. L4 / 5 facet articular cartilage specimens were collected from patients undergoing lumbar surgery. The cartilage structure was observed by hematoxylin-eosin staining, Phanerin O staining, toluidine blue staining and Masson staining. Results and conclusion: hematoxylin-eosin staining. All layers of articular cartilage could be clearly observed by phenyraniline-solid green staining and toluidine blue staining, among which hematoxylin and eosin staining could clearly display the chondrocytes, tidal lines and subchondral bones. The nuclei of chondrocytes were blue-purple. The four layers of cartilage were clearly shown by the staining of reddish and solid green: superficial layer (surface of cartilage, middle layer (round cells, light staining, regular arrangement), columnar cell layer (larger cells). Multinucleate, arranged neatly, showed that the cartilage matrix was even light red, the subchondral bone was green, the cartilage tissue and bone tissue were contrasting distinctly. Toluidine blue staining showed that the tissue structure layer was not clear and the cell nucleus staining was clear. The cytoplasm almost did not stain, the matrix was lavender and purple, but the demarcation between the cartilage layer and the subchondral bone layer was clear, the cartilage layer was homogeneous and dark red, and the cell staining was light. There was no obvious boundary between the matrix and the cartilage. Masson staining was clear for collagen fibers. The results showed that the structure of cartilage and subchondral bone was the best in all layers of cartilage and subchondral bone, and the results showed that the structure of cartilage and subchondral bone was the best. The morphological changes of chondrocytes were observed by hematoxylin-eosin staining.
