X线辐射后A549细胞上清外泌体GPC1表达与细胞周期的关系

【摘要】:目的观察X线辐射对A549肺腺癌细胞周期及对细胞上清外泌体磷脂酰肌醇蛋白聚糖(GPC)1表达的影响。方法培养肺腺癌细胞A549并分别给予0、0.5、1、1.5、2 Gy剂量照射,照射后继续培养6、12 h收集细胞,用PI法检测细胞周期;收集细胞上清液,超速离心法提取外泌体,Western印迹检测外泌体GPC1表达。结果 G_2期细胞的比例随着剂量的增加而增加,差异有统计学意义(P0.001),其他各剂量组与2 Gy组比较差异有统计学意义(P0.05);各剂量组S期细胞比例均高于0 Gy组(P0.05);Western印迹结果显示,随辐射剂量增大,GPC1表达量逐渐降低。结论辐射能够诱导A549细胞G2期、S期阻滞,同时细胞上清外泌体GPC1蛋白表达减少,二者具有相关性。
[Abstract]:Objective to observe the effects of X-ray radiation on the cell cycle and the expression of phosphatidylinositol proteoglycan 1 in the supernatant of A549 lung adenocarcinoma cells. The cell cycle was detected by Pi assay, the supernatant was collected and the exocrine GPC1 expression was detected by Western blotting with ultracentrifugation. The difference was statistically significant (P 0.001), and there was significant difference between other dose groups and 2 Gy groups (P 0.05), and the percentage of S phase cells in each dose group was higher than that in 0 Gy group (P 0.05), and the results of Western blotting showed that the percentage of S phase cells in each dose group was higher than that in 0 Gy group. Conclusion radiation can induce G 2 phase arrest of A549 cells and decrease the expression of GPC1 protein in the exocrine of the cell supernatant. There is a correlation between the expression of GPC1 and the expression of GPC1 protein in A549 cells.
