超高效液相色谱-四极杆飞行时间质谱联用技术分析人类唾液代谢组的性别依赖性

【摘要】:目的·研究人类唾液代谢组的性别依赖性。方法·使用基于超高效液相色谱 四极杆飞行时间质谱(UHPLC-Q-TOFMS)联用的代谢组学技术,检测分析年龄、体质量指数、生活及洁牙条件匹配的5位男性与5位女性健康志愿者的唾液样本代谢物组成,再通过与已有数据库对比和标准品验证鉴定唾液中的代谢物群,确定2组样品中差异具有统计学意义的代谢物。结果·鉴定了人类唾液中的13种氨基酸、6种胆碱代谢物、15种肉碱类代谢物、4种鞘氨醇类代谢物、7种溶血磷脂酰胆碱和3种有机酸等48种代谢物。发现苯丙氨酸、乙酰基肉碱、丙酰基肉碱、丁酰基肉碱、异丁酰基肉碱、异戊酰基肉碱和鞘氨醇的含量在女性唾液中显著高于男性样本。结论·人类唾液中含有丰富的代谢物信息,唾液代谢组研究必须考虑其性别依赖性。
[Abstract]:Objective to study the sex dependence of human saliva metabolism.Methods Metabolomics technique based on UHPLC-Q-TOFMS was used to detect and analyze age and body mass index.The metabolites in saliva samples of 5 male and 5 female healthy volunteers matched with the living and cleaning conditions were identified by comparing with existing databases and identifying the metabolites in saliva.The metabolites with statistical significance were determined in the two groups.Results 13 kinds of amino acids, 6 kinds of choline metabolites, 15 kinds of carnitine metabolites, 4 kinds of sphingosine metabolites, 7 kinds of lysophosphatidylcholine and 3 kinds of organic acids in human saliva were identified.It was found that the contents of phenylalanine, acetyl carnitine, propionyl carnitine, butyryl carnitine, isobutylol carnitine, isovalerate carnitine and sphingosine were significantly higher in female saliva than in male sample.Conclusion there are abundant metabolites in human saliva, and its sex dependence must be considered in the study of salivary metabolites.
