输注聚合人胎盘血红蛋白对大鼠氧化损伤的初步研究

【摘要】:目的探讨输注不同浓度聚合人胎盘血红蛋白(Poly PHb)对大鼠造成的氧化损伤。方法建立动物换血模型,将50只SD大鼠随机均分为假手术组:仅做股动/静脉插管处理;氯化钠注射液(对照)组:股动/静脉插管,输注氯化钠注射液行换血实验;2%、4%及6%Poly PHb 3个实验组:股动/静脉插管,分别输注相应浓度的Poly PHb行换血实验。分别在大鼠换血前、换血70%后即刻观测平均动脉压(MAP),换血完成后即刻从大鼠股动脉取血0.5 m L/只,做动脉血血气分析,并检测各组大鼠血浆中还原型谷胱甘肽(GSH)浓度、超氧化物歧化酶(SOD)活力、总抗氧化能力(T-AOC)、丙二醛(MDA)浓度、8-羟基脱氧鸟苷(8-OHDG)浓度。结果 1)大鼠换血70%后,2%、4%及6%Poly PHb组与对照组相比,GSH(μmol/L)分别为33.76±14.33、47.98±21.88、39.73±15.54及10.25±5.70(P0.01);SOD(U/m L)分别为238.07±27.39、355.73±35.42、405.26±45.01及138.46±57.16(P0.01);T-AOC(U/m L)分别为20.50±7.87、32.65±4.38、38.03±4.50及5.40±2.22(P0.01)。2)大鼠换血70%后,2%、4%及6%Poly PHb组与对照组相比,MDA浓度(nmol/m L)随着Poly PHb浓度的增高而增高:45.08±4.12、54.39±3.16、58.52±6.33及2.85±0.59(P0.01)。3)大鼠换血70%后,2%、4%及6%Poly PHb组与对照组相比,大鼠血浆8-OHDG浓度(nmol/m L)未发生明显变化:2.54!0.90、2.76!0.99、2.79!0.82及2.26!0.42(P0.05)。结论 Poly PHb对大鼠造成脂质过氧化损伤,且氧化损伤程度随着Poly PHb浓度的增高而加重,但并未对大鼠DNA造成损伤。
[Abstract]:Objective to investigate the oxidative damage induced by different concentrations of polymerized human placental hemoglobin (Poly PHb) in rats.Methods 50 SD rats were randomly divided into sham-operation group (femoral artery / vein intubation only) and sodium chloride injection (control) group (femoral / venous intubation).Three experimental groups: femoral artery / vein catheterization, transfusion of sodium chloride injection with 4% and 6%Poly PHb, and transfusion of corresponding concentration of Poly PHb were performed.The mean arterial pressure (MAPH) was measured immediately before and 70% after the exchange of blood in rats, and 0.5 mL of blood was taken from the femoral artery of rats immediately after the exchange of blood.
