【摘要】:目的建立一种基于时间分辨荧光纳米颗粒的磁分离均相免疫分析方法并用于检测降钙素原(PCT)重组抗原。方法以PCT单克隆抗体偶联铕离子荧光微球为检测抗体,另一株PCT单克隆抗体偶联生物素作为固相抗体,建立一种通过表面修饰有链霉亲和素的磁珠分离铕离子荧光微球-检测抗体-降钙素原-固相抗体-生物素复合物的均相免疫分析方法。用该法检测PCT重组抗原,通过Sigma Plot分析该法的最低检测限。结果 PCT重组抗原的标准曲线匹配二次函数,函数方程为Y=19170.12+75493.74X-26.00X2(r=0.9986),PCT重组抗原检测灵敏度为0.04 ng/ml。结论铕离子荧光微球可作为标记物应用于磁分离均相免疫分析方法,并在PCT重组抗原检测中获得较高的检测灵敏度。
[Abstract]:Objective to establish a time resolved fluorescent nanoparticles based magnetic separation homogeneous immunoassay method for the detection of procalcitonin PCT recombinant antigen. Methods PCT monoclonal antibody coupled with europium ion fluorescent microspheres was used as detection antibody and another strain of PCT monoclonal antibody coupled with biotin as solid phase antibody. A homogeneous immunoassay method was developed for the separation of europium ion fluorescent microspheres by magnetic beads modified with streptavidin on the surface-detection antibody-procalcitonin-solid phase antibody-biotin complex. PCT recombinant antigen was detected by this method, and the minimum detection limit was analyzed by Sigma blot. Results the standard curve matching quadratic function of PCT recombinant antigen was used. The function equation was: YT19170.12 75493.74X-26.00X2RX 0.9986. The detection sensitivity of PCT recombinant antigen was 0.04 ng / ml. Conclusion europium ion fluorescent microspheres can be used as markers for magnetic separation homogenous immunoassay, and high sensitivity can be obtained in the detection of PCT recombinant antigen.
