重庆地区汉族人群KIR基因多态性研究

【摘要】:目的研究重庆地区汉族人群KIR基因及基因型多态性特点。方法采集316名重庆地区汉族无偿献血者的外周血样,应用多重PCR-SSP方法作KIR基因分型;根据等位基因频率网络数据库(AFND)中的ID号命名检测到的基因型及单体型组成,以Spss17.0软件对人群间KIR基因频率作卡方检验。结果 316名重庆地区汉族献血者个体均含有KIR基因3DL2、3DL3;2DL4、2DL1、3DP1、2DP1、2DL3、3DL1较为常见,频率分别为94.1%、92.0%、92.0%、90.3%、87.4%、76.8%;频率较低(30%)的基因有2DL5、2DS1、3DS1、2DS5、2DL2、2DS2和2DS3。共发现24种KIR基因型,其中以ID号为1、2和8的基因型最为常见,频率分别为54.4%(172/316)、15.2%(48/316)和6.3%(20/316),还有1种基因型无法在AFND中查出其基因型。重庆地区汉族献血者携带的KIR各基因中分别有8个和7个基因的频率与南非圣族人和高加索人明显不同(P0.05),有5个与乌市维族人明显不同(P0.05),仅有1个与拉萨藏族有明显不同(P0.05),而与四川汉族、云南汉族、浙江汉族、江苏汉族和韩国人均无明显差异(P0.05)。结论重庆地区汉族人群的KIR基因分析既含丰富的多态性,又具有自身的特点,与四川、云南等中国南方汉族人群一致。
[Abstract]:Objective to study the polymorphism of KIR gene and genotype in Chongqing Han population. Methods Peripheral blood samples were collected from 316 non-remunerated blood donors of Han nationality in Chongqing, and the genotypes and haplotypes were detected according to the ID number of the allele frequency network database (AFND), and the multiple PCR-SSP method was used for KIR genotyping. The frequency of KIR gene was detected by chi-square with Spss17.0 software. Results the 3DL2DL3DL3DL3DL4DL4DL4DL2DL2DL2DL3DL3DL2DL2DL3DL1 gene 3DL2DL2DL2DL3DL1 was found in 316 blood donors of Han nationality in Chongqing. The frequencies of 3DL2DL1DL3DL3DL2D2D2D2D2DL2D2D2DL2D2DS2DS2DS2DS2DS2DS2and 2DS3D3D2DS2DS2DS2DS2DS2DS2DS2DS2DS2DS2D2DS2DS2DS2DS2DS2DS2DS2DS2DS2DS2DS2DS2DS2DS2DS2DS2DS2@@ A total of 24 genotypes of KIR were found, of which ID number 1G 2 and genotype 8 were the most common, with frequencies of 54.4% (17.2 / 316) and 6.3% (20 / 316), respectively. One genotype could not be detected in AFND. The frequencies of 8 and 7 of the KIR genes carried by Han blood donors in Chongqing area were significantly different from those of the South African saints and Caucasians (P0.05), and 5 of them were significantly different from those of Uighurs in Wushi (P0.05), and only one of them had the same frequency with Lhasa Tibetans. The difference was significant (P0.05), but it was different from that of the Han nationality in Sichuan. There was no significant difference between Han people in Yunnan, Zhejiang, Jiangsu and Korea (P0.05). Conclusion the KIR gene analysis of Han population in Chongqing area is not only rich in polymorphism, but also has its own characteristics, which is consistent with the Han population in southern China, such as Sichuan and Yunnan.
