富含细胞因子自体血清的制备及细胞因子分析

【摘要】：背景:自体血清疗法是一种新兴的、安全的生物疗法,主要适应证是骨关节炎、风湿性关节炎等类似病症。目的:探讨富含细胞因子的自体血清制备方法并进行细胞因子分析。方法:采用了获得专利的采血设备(内含3 mm玻璃小珠)进行外周血采集,充分混匀后放置于37℃的恒温培养箱内,进行24 h的恒温孵育,最终分离过滤得到自体血清。测定白细胞介素1受体拮抗物、白细胞介素6、白细胞介素8、白细胞介素1β等细胞因子定量的浓度,并用细胞因子抗体芯片的检测方法对36种细胞因子进行了半定量测定。结果与结论:(1)制备的自体血清中多种细胞因子浓度有明显增加,其中,对缓解骨关节炎病症起到关键作用的白细胞介素1受体拮抗物的质量浓度由平均140.25 ng/L增加至1 125 ng/L,增加8倍以上;白细胞介素6质量浓度由389.5 ng/L增加至2 802 ng/L,白细胞介素8由"未检测到"增加至2 822 ng/L;(2)采用细胞因子抗体芯片的方法检测制备的自体血清发现,至少有10种细胞因子的含量发生了改变,大部分因子的变化趋势是制备后增加,少数几个因子呈下降的趋势;(3)结果证明,制备的自体血清当中,含有大量的白细胞介素1受体拮抗物和多种细胞因子,另外,自体血清中还含有一部分的趋化因子,提示这些趋化因子能够招募更多的免疫细胞来到炎症部位,加快炎症反应进程,起到治疗的作用。
