皮肤肌肉切口牵拉术对幼年大鼠成年后空间学习记忆能力的影响

【摘要】:目的观察皮肤肌肉切口牵拉术对幼年大鼠机械痛阈和成年后空间学习记忆能力的影响。方法 3周龄幼年大鼠27只,体重60~80 g,随机分成3组(n=9):空白对照组(C组)、假手术组(Sham组)、皮肤/肌肉切口牵拉模型组(skin/muscle incision and retraction,SMIR组)。SMIR组行皮肤肌肉切口牵拉手术,Sham组行肌肉切开未作牵开处理,C组大鼠不进行手术操作。3组大鼠分别在术前、术后1、3、7、12、22、32 d时,测定机械痛阈(MWT)。各时间点测试完毕后,3组大鼠于术后33 d行Morris水迷宫实验。结果 SMIR组大鼠术后1天痛阈降低(P0.05),3组大鼠在术前及术后3、7、12、22、32 d痛域阈值差异无统计学意义(P0.05)。Morris水迷宫实验中,与Sham组和C组相比,SMIR组Morris水迷宫定向航行实验平均潜伏期延长(P0.01);SMIR组大鼠在平台所在象限的游泳时间和路径与总游泳时间和路径的比例显著减小(P0.01);Sham组与C组间差异均无统计学意义(P0.05)。结论幼年大鼠SMIR术不引起慢性疼痛,而损害空间学习和记忆功能。
[Abstract]:Objective to observe the effect of skin muscle incision traction on mechanical pain threshold and spatial learning and memory ability in juvenile rats. Methods 27 3-week-old young rats were used. Weight 60,80g, randomly divided into 3 groups: blank control group (group C), sham operation group (Sham group), skin / muscle incision pulling model group (skin/muscle incision and retraction,SMIR group). The rats of group 3 were not operated on before operation. Mechanical pain threshold (MWT).) was measured at 1: 3, 7, 12, 22 and 32 days postoperatively. The Morris water maze test was performed 33 days after each time point test. Results there was no significant difference in pain threshold between the 3 groups before and after operation (P0.05). The pain threshold of the SMIR group was not significantly different from that of the control group (P0.05). Morris water labyrinth test showed that there was no significant difference in the pain threshold between the three groups before and after operation (P0.05). Compared with Sham group and C group, the average latency of Morris water maze directional navigation experiment in SMIR group was prolonged (P0.01) and the proportion of swimming time and path to total swimming time and path in platform quadrant decreased significantly (P0.01) and the difference between group C and group C was significantly decreased (P0.01). There was no statistical significance (P0.05). Conclusion SMIR does not cause chronic pain, but impairs spatial learning and memory in juvenile rats.
