正常睡眠觉醒周期及睡眠剥夺条件下内侧前额叶皮层神经元自噬活动特征及机制研究

【摘要】:目的观察睡眠剥夺(sleep deprivation,SD)对内侧前额叶皮层(medial prefrontal cortex,m PFC)神经元自噬(autophagy)活动的影响及机制。方法实验使用成年(8~10周)雄性C57BL/6小鼠,体重23-26g,随机分为睡眠剥夺6 h(SD6 h)组与其对照组(n=10-12只/组)、睡眠剥夺12 h(SD12 h)组与其对照组(n=10-12只/组)。采用轻柔刺激法建立SD6 h和SD12 h模型。蛋白免疫印迹(Western blot)检测内侧前额叶神经元皮层超氧化物歧化酶2(superoxide dismutase,SOD2)、过氧化氢酶(catalase)、沉默信息调节因子3(Silent information regulator 3,Sirt3)、微管相关蛋白1轻链3-Ⅱ(LC3-Ⅱ)、P62(也称SQSTM1)、Beclin-1、腺苷酸活化蛋白激酶(AMP-activated protein kinase,AMPK)、雷帕霉素靶蛋白(mammalian target of rapamycin,m TOR)、p-AMPK、p-m TOR蛋白的表达水平。免疫荧光染色观察内侧前额叶皮层神经元溶酶体膜相关蛋白1(lysosomal-associated membrane protein 1,LAMP-1)的表达情况,Dihydroethidine(DHE)染色检测m PFC神经元O2-的水平。结果1.在正常睡眠觉醒周期中,与白天相比,夜间自噬流活性较高,P62蛋白表达较低,Beclin-1蛋白表达相对较高。2.与对照组相比,SD6 h后m PFC神经元SOD2(P0.05)、Catalase(P0.05)和Sirt3(P0.05)蛋白表达均显著上调,O2-的水平同时也相对升高(P0.05);而SD12 h后,m PFC神经元SOD2、Catalase和Sirt3蛋白表达水平与对照组相比均无显著差异,但O2-的水平相对升高。3.与对照组相比,SD6 h与SD12 h均使m PFC神经元LC3-Ⅱ(P0.05)和P62的表达水平升高(P0.05),进一步研究发现,Beclin-1(P0.05)和LAMP-1(P0.05)的表达水平均相对升高。4.SD6 h后,与对照组相比,m PFC神经元AMPK和m TOR蛋白水平无显著变化;SD12 h后,与对照组相比,m PFC神经元m TOR蛋白表达水平显著下调(P0.05),但AMPK蛋白表达无明显变化。结论1.睡眠期间,m PFC神经元自噬活动较低,而觉醒期间m PFC神经元自噬活动较高。2.睡眠剥夺后,m PFC神经元氧化水平升高,而抗氧化水平只产生一过性升高。3.无论是SD6 h还是SD12 h,均使m PFC神经元自噬活动增强。4.SD12 h使m PFC神经元m TOR信号通路被抑制,进而诱导自噬活动上调。
