伏立康唑对体外培养棘阿米巴增殖和形态表现的影响

【摘要】:目的:探讨伏立康唑对体外培养棘阿米巴的增殖和形态表现的影响,阐明伏立康唑对棘阿米巴滋养体和包囊的抑制和杀伤作用。方法:选取对数生长期的棘阿米巴原虫分为对照组和实验组(2.5和25.0mg·L~(-1)),分别于药物作用后24、48、72和96h收集各组虫体,计数虫体数量并绘制增殖曲线;光镜下观察棘阿米巴的形态表现、活力和贴壁情况;电镜下观察其超微结构。结果:与对照组比较,实验组棘阿米巴虫体数量减少(P0.01);光镜下实验组棘阿米巴形态变化明显,由不规则带棘状伪足的滋养体转化为圆形包囊,出现大量细胞碎片;电镜下实验组棘阿米巴的超微结构出现不同程度的破坏、坏死。结论:一定浓度伏立康唑能够有效抑制体外培养棘阿米巴的增殖并改变其形态甚至超微结构,对滋养体和包囊均具有明显杀伤作用。
[Abstract]:Aim: to investigate the effect of vericonazole on proliferation and morphology of cultured Acanthamoeba in vitro, and to elucidate the inhibitory and killing effects of voriconazole on trophozoites and cysts of Acanthopsis amoeba. Methods: the protozoa of Acanthoamoeba in logarithmic growth stage were divided into two groups: control group and experimental group (2.5 and 25.0mg L ~ (-1), were collected at 2448 / 72 and 96 hours after treatment, respectively), the number of parasites was counted and the proliferation curve was plotted. The morphology, activity and adherent of Acanthamoeba were observed under light microscope, and ultrastructure was observed under electron microscope. Results: compared with the control group, the number of Acanthoamoeba in the experimental group was decreased (P0.01), the morphological changes of Acanthamoeba in the experimental group were obvious under light microscope, the trophozoites with irregular and spinous pseudopodium were transformed into circular cysts, and a large number of cell fragments appeared. The ultrastructure of Acanthamoeba in the experimental group was destroyed and necrosed in different degree under electron microscope. Conclusion: Veconazole at a certain concentration can effectively inhibit the proliferation of Acanthamoeba in vitro and change its morphology and even ultrastructure. It can kill trophozoites and cysts.
