极化液对内毒素休克兔脑保护作用的研究
[Abstract]:Objective: to study the protective effect of polarizing liquid on the brain of rabbits with endotoxic shock and to explore its possible mechanism. Methods: thirty New Zealand white rabbits were randomly divided into three groups: normal control group (NS group), endotoxin shock group (ES group), polarized solution group (GIK group), 10 rabbits in each group. NS group received intravenous infusion of compound sodium chloride (5ml/kg.h). The endotoxic shock model was established by intravenous injection of LPS 0.5mg/kg in ES group and GIK group, and then was treated with compound sodium chloride and GIK (5ml/kg.h for 6 hours). The vital signs (mean arterial pressure, respiratory rate, body temperature), blood glucose and blood gas analysis indexes (PH,PaCO_2, blood potassium, lactic acid), ELISA method) were measured at each time point. Serum NSE concentration was measured by lactic acid), ELISA method. At the end of the experiment, the white rabbits were killed and 10% tissue homogenate was prepared in the left cerebral cortex. The oxidation indexes (H 2O 2, no, iNOS, MDA), antioxidant index (SOD,T-AOC) were measured. The activities of Na~-K- ATP enzyme and Ca~ (2)-Mg~ (2)-ATP enzyme were measured, the dry and wet weight of the remaining brain tissue was measured and the water content was calculated. Morphological changes of right cerebral cortex were observed under light and electron microscope. Results: 1. Compared with NS group, the mean arterial pressure (map), PH and PaCO_2 (P < 0.05) and respiratory rate (P < 0.05) were significantly decreased after LPS injection in the experimental rabbits. The mean arterial pressure and lactic acid increased gradually during the treatment in). ES group and GIK group, and the mean arterial pressure increased more significantly in GIK group than that in ES group (P0. 05), while the mean arterial pressure and lactic acid in GIK group were significantly higher than those in ES group (P < 0. 05). Lactic acid decreased significantly (P < 0.05), but there was no statistical difference between the other indexes (P0.05). Compared with NS group, ES group and GIK group, the oxidative index in brain homogenate, serum NSE concentration and brain water content were significantly increased (P < 0.05), and the antioxidant index in homogenate was significantly higher than that in GIK group (P < 0.05). The activities of Na~-K- ATP enzyme and Ca~ (2)-Mg~ (2)-ATP enzyme were significantly decreased (P0. 05). Compared with ES group, the oxidative index, NSE concentration and brain water content in GIK group were significantly decreased, while the antioxidant index, Na~-K3. ATP enzyme and Ca~ (2)-Mg~ (2)-ATP enzyme activity were significantly increased (P < 0. 05). No abnormality was observed in NS group. The damage of brain cells and mitochondria in ES group and GIK group was obvious, but the pathological damage in GIK group was lighter than that in ES group. Conclusion: polarizing solution has protective effect on brain injury in rabbits with endotoxic shock, which may be related to the improvement of hemodynamics, reduction of oxidative stress level and reduction of mitochondrial damage.
