中空介孔普鲁士蓝纳米粒多模态成像及增效HIFU/化疗协同治疗的实验研究

第一部分中空介孔普鲁士蓝纳米粒的制备、表征及释药研究目的制备了一种具有优异生物相容性/稳定性的多功能纳米粒——装载温敏性相变材料全氟己烷和抗肿瘤药物阿霉素的中空介孔普鲁士蓝纳米材料(perfluorohexane and the doxorubicin-encapsulated hollow mesoporous Prussian Blue nanoparticles,HMPBs-DOX/PFH),检测其物理性质、化学性质、光学性质、稳定性、药物释放性能以及相变性能。方法采用选择性刻蚀法和药物吸附法依次制备出HMPBs和HMPBs-DOX,检测其形态结构及化学组成。然后,采用真空灌注法制备出HMPBs-DOX/PFH,对其粒径、电位、载药率以及光吸收性等性能。通过光学显微镜分别检测该多功能纳米粒的相变效应。检测HMPBs-DOX/PFH分别在不同p H值(4.6,6.0,7.4)缓冲液和HIFU作用前后的药物释放率,并检测生物组织中HMPBs-DOX/PFH在HIFU作用前后的药物释放情况。最后采用高效液相色谱法检测DOX的热稳定性。将HMPBs-DOX/PFH与MDA-MB231细胞...
