三个STR基因座荧光标记复合扩增的研究及其法医学应用

【摘要】:目的 本课题旨在寻找新的有应用价值的STR基因座,并建立D12S1064、GATA140E03、D18S536基因座的荧光标记复合扩增体系。按照美国DNA分析方法技术工作组(TWGDAM)的指导方案,进行了法医学应用性研究。方法 首先对5个新的STR基因座,即D9S1119、D12S1064、D18S536、GATA156D11和GATA140E03,进行群体遗传学研究。从中获得三个多态性好、个人识别机率高的三个基因座,,即D12S1064、GATA140E03、D18S536,利用传统的复合扩增的方法,用美国ABI-310毛细管电泳仪进行荧光标记复合扩增的研究。并对复合扩增系统的灵敏度和准确度,基因座的种属特异性,不同退火温度下复合扩增的效果等进行了重复性研究。结果 获得了5个STR基因座的基因频率、个人识别率、非父排除率等法医鉴定中所需要的遗传学数据;成功建立了D12S1064、GATA140E03、
[Abstract]:Objective to search for a new STR locus and to establish a fluorescent multiplex amplification system for D12S1064-GATA140E0E03 D18S536 locus. The application of forensic science was studied according to the guidance of (TWGDAM) of the DNA Analytical Methodology Technical working Group in the United States. Methods five new STR loci, D9S1119, D12S1064, D18S536, GATA156D11 and GATA140E03, were studied in population genetics. Three loci, D12S1064, GATA140E03, D18S536, which have good polymorphism and high probability of personal identification, were obtained. The fluorescence labeling multiplex amplification was carried out by ABI-310 capillary electrophoresis in the United States. The sensitivity and accuracy of the multiplex amplification system, the specificity of gene loci and the effect of multiple amplification at different annealing temperatures were studied repeatedly. Results genetic data of 5 STR loci such as gene frequency, personal identification rate, non-paternal exclusion rate and so on, were obtained, and D12S1064-GATA140E03 was successfully established.
