基于mRNA稳定性推断死亡时间的研究

【摘要】:目的探讨大鼠死后不同时间肾、脾3-磷酸甘油醛脱氢酶(GAPDH)mRNA的表达对死亡时间(PMI)推断的应用价值。方法28只大鼠断颈处死后置于20℃温控系统内,分别于死后0,8,16,24,32,40,48h共7个时相点(n=4)解剖,利用荧光RT-PCR技术检测大鼠肾、脾GAPDHmRNA相对表达量的变化。结果在大鼠死后40h肾、32h脾内可检测出GAPDHmRNA,且其表达产物均呈规律性下降趋势(荧光强度逐渐变暗并最终消失)。结论检测大鼠死后不同时间肾、脾GAPDHmRNA的表达对PMI推断具有一定的应用价值。
