大鼠心肌和骨骼肌肌钙蛋白I降解与死亡时间关系的研究

【摘要】: 目的 研究大鼠心肌肌钙蛋白I(Cardiac troponin I, cTnI)和骨骼肌肌钙蛋白I(Skeletal troponin I, sTnI)在体和离体两种情况下的降解规律并进行比较,探讨根据cTnI及sTnI降解情况推断死亡时间的可行性,为死亡时间(Postmortem interval,PMI)推断的研究提供新的思路和方法,为法医实践中PMI的推断提供科学依据。 方法 将健康SD大鼠10只按在体和离体二种处置方式随机平均分为2组,每组5只。将10只大鼠用机械性窒息法致死,取离体组大鼠心肌及骨骼肌组织置于保湿盒内与在体组大鼠尸体一起放入人工气候箱中,恒温30℃,湿度恒定为40%保存。分别于取材后0、24、48、72、96和120h提取离体及在体组织中的蛋白质,应用免疫印迹(Western blot)法结合图像分析技术半定量检测当时死亡及离体时间的大鼠cTnI、sTnI含量。用死后120h时cTnI或sTnI的含量占死亡即刻cTnI或sTnI的含量的百分数来表示cTnI及sTnI降解的相对速度。将所得数据进行统计学分析,寻找随着PMI的延长各含量参数的变化规律,计算各参数与PMI的相关系数,求出回归方程。最后根据分析结果比较在体和离体cTnI或sTnI降解规律之间的异同。 结果 1、在体及离体情况下大鼠cTnI与sTnI的含量均随死亡或离体时间延长而逐渐下降,且与死亡或离体时间的对数值有很强的线性关系。 2、在体情况下cTnI与sTnI的降解比离体情况慢。 3、在体和离体两种情况下sTnI均比cTnI降解慢。 结论 1、cTnI和sTnI有望用于推断五天以内的死后经过时间。 2、cTnI和sTnI在离体与在体条件下的降解情况并不完全一致,离体时的降解速度要比在体时快。 3、sTnI较cTnI更适用于推断PMI。
