小鼠皮肤切创愈合过程中caspase-8表达及其与时间相关性的研究

【摘要】:前言 皮肤损伤愈合的过程可分为三个阶段,即炎症期、增生期及组织重建期。中性粒细胞和巨噬细胞的渗出是该过程重要的防御机制,它们吞噬并消化细菌等外源性微粒状物质及死亡细胞碎片。许多研究表明在这些细胞数量减少的同时有细胞的凋亡。 细胞的程序性死亡也叫做细胞凋亡,是细胞自我毁坏的生理过程。凋亡在生物发育、维持多细胞生物体内环境稳定及宿主防御中起十分重要的作用。细胞凋亡的失调涉及癌症、自身免疫疾病、神经变性疾病及局部缺血损伤等多种疾病。 caspase是诱导细胞凋亡的重要的蛋白酶,有14个家族成员:caspase-1~-14。caspase既是半胱氨酸蛋白酶能以半胱氨酸为亲核基团裂解底物,又是天冬氨酸蛋白酶可将天冬氨酸C-端肽链酶切。caspase家族的反应是级联放大的。上游启动型caspase(如caspase-8、-9)被caspase适配器活化后将激活效应型caspase(如caspase-3、-6、-7)使细胞蛋白裂解,导致细胞凋亡。启动型caspase的活化是细胞凋亡的调控点。 作为caspase家族的启动因子,caspase-8在皮肤损伤愈合过程中起重要作用。本研究用免疫组织化学染色技术和Western blot方法检测小鼠皮肤切创愈合过程中caspase-8在多核粒细胞、单核细胞及成纤维细胞中的表达并探讨通过检测caspase-8表达的规律推测皮肤损伤时间的可行性。 材料和方法 1.动物模型建立及分组 健康成年清洁级昆明系雄性小鼠33只,体重35g-40g,随机分成11
[Abstract]:The healing process of skin injury can be divided into three stages: inflammation stage, proliferative stage and tissue reconstruction stage. The exudation of neutrophils and macrophages is an important defense mechanism in this process. They engulf and digest foreign particulate matter such as bacteria and death cell fragments. Many studies have shown that the number of these cells decreases at the same time as cell apoptosis. Programmed cell death, also known as apoptosis, is a physiological process of cell self-destruction. Apoptosis plays an important role in biological development, maintenance of environmental stability in multicellular organisms and host defense. Apoptosis disorders are associated with cancer, autoimmune diseases, neurodegenerative diseases and ischemic damage. Caspase is an important protease that induces cell apoptosis. There are 14 family members: caspase-1~-14.caspase is a cysteine protease that can use cysteine as the nucleophilic group lysate substrate. Aspartate protease can cut the C-terminal peptide chain of aspartic acid. The reaction of caspase family is amplified by cascade. When upstream caspase (such as caspase-8,-9) is activated by caspase adaptor, the activated caspase (such as caspase-3,-6,-7) cleavage the cell protein and induce cell apoptosis. Activation of priming caspase is the regulatory point of apoptosis. As the promoter of caspase family, caspase-8 plays an important role in the healing of skin injury. In this study, immunohistochemical staining and Western blot methods were used to detect caspase-8 in polymorphonuclear granulocytes (PMN) during skin wound healing in mice. The expression of caspase-8 in monocytes and fibroblasts and the feasibility of estimating the time of skin injury by detecting the expression of caspase-8. Materials and methods 1.
