ABO基因分型及其法庭科学应用研究

【摘要】:目的 为使ABO基因型更好地服务于法庭科学实践,本研究建立和优化ABO基因分型方法,并充分评估ABO基因型在法庭科学实践中的可信性及应用价值。方法 采用PCR-RFLP技术,复合扩增产物用Kpn Ⅰ和Alu Ⅰ限制性内切酶酶切,经聚丙烯酰胺凝胶电泳及硝酸银染色判读ABO基因型。运用该方法对血样、混合斑、唾液斑、毛发、肌肉、骨等常见生物学样本进行ABO基因分型。用PCR-直接测序法对ABO基因进行序列分析。结果 该方法将ABO血型直接区分为常见的六种基因型。两代家系的ABO基因型结果均符合孟德尔遗传规律。同一个体的ABO基因型与ABO表型结果完全相符。100例认定嫌疑人的强奸案中混合斑精子的ABO基因型与相应嫌疑人血样的ABO基因型结果完全一致。常见生物学检材均能得到准确的ABO基因型检测结果。经PCR-直接测序法检出一例ABO基因突变。结论 运用PCR-RFLP
