长春汉族群体D1S549等七个短串联重复序列基因座的遗传多态性研究

【摘要】: STR是一类具有长度多态性的DNA序列,按照孟德尔共显性方式遗传,故已被作为遗传标记广泛应用在生物科学和医学等领域的研究与实践当中。STR的应用非常广泛,特别是STR基因座的PCR扩增片段长度多态性检测技术,该技术有着十分显著的适用于法医学个人识别的特点。本研究选择了D1S549,D3S1744, D6S366,D5S818、D8S1179、D13S317、D16S539等7个STR基因座,采用PCR、DNA聚丙烯酰胺凝胶电泳银染分型技术对长春汉族群体这7个位点的基因频率分布进行了调查,得出此7个基因座在长春汉族人群中基因型频率分布符合Hardy-Weinberg平衡,提供的信息量大,有较高的个人识别能力和非父排除概率。除D6S366位点杂合度为0.6333,小于0.8,其余几个基因座的杂合度均在0.8以上而多态信息含量均在0.7以上,属多态信息含量较高的基因座,个人识别能力除D6S366略低(0.7793),其余都在0.9以上,非父排除概率除D3S1744和D6S366略低(分别为0.4184和0.3617),其余均在0.6以上,属实用价值比较高的基因座。
[Abstract]:STR is a kind of DNA sequence with length polymorphism, which is inherited according to Mendelian codominant mode, so it has been widely used as genetic marker in the research and practice of bioscience and medicine. In particular, the PCR amplified fragment length polymorphism detection technique of STR loci is very suitable for forensic personal recognition. Seven STR loci D1S549, D3S1744, D6S366, D5S818, D8S1179, D13S317, D16S539 were selected. PCR,DNA polyacrylamide gel electrophoresis silver staining technique was used to investigate the gene frequency distribution of these seven loci in Changchun Han population. It is concluded that the genotypic frequency distribution of these seven loci in Changchun Han population accords with Hardy-Weinberg balance, provides a large amount of information, and has high personal recognition ability and non-paternity exclusion probability. Except that the heterozygosity of D6S366 locus is 0.6333, which is less than 0.8, the heterozygosity of the other loci is above 0.8, and the polymorphism information content is above 0.7. it belongs to the loci with high polymorphism information content. The personal recognition ability was slightly lower (0.7793) than that of D6S366, and the probability of non-paternal exclusion was slightly lower than that of D3S1744 and D6S366 (0.4184 and 0.3617, respectively), which belonged to loci with high practical value.
