紫苏籽油及原花青素的提取工艺研究

【摘要】:紫苏是我国传统的中药材和油料作物,紫苏籽含油量高,皮中含有原花青素,对紫苏籽进行综合利用市场前景广阔。本文以紫苏籽为原料,采用超声波辅助水酶法提取紫苏籽油,优化工艺,并测定紫苏籽油的性质;同时研究了紫苏籽皮原花青素的提取及纯化工艺,并对其抗氧化活性进行了测定。主要研究结果如下:(1)在单因素试验基础上,通过响应面法得到超声波辅助水酶法提取紫苏籽油的最优条件:复合酶配比(纤维素酶用量5.5%、中性蛋白酶用量4.5%、果胶酶用量7.5%),液料比6:1(m L/g),超声时间23min,酶解温度50℃,酶解时间2.5h,出油率为30.45%。(2)对水酶法、热榨法、冷榨法及有机溶剂浸提法所提取的紫苏籽油的主要理化性质、脂肪酸组成、生物活性成分含量以及抗氧化能力进行了测定与比较,结果表明:水酶法油的过氧化值最低,碘值最高;水酶法油的棕榈酸含量低,油酸含量显著高于热榨油与有机溶剂浸提油,α-亚麻酸与不饱和脂肪酸含量最高;水酶法油的VE含量与有机溶剂浸提油相差不大,多酚、总黄酮及甾醇含量最高;通过DPPH法、ABTS法和FRAP法得出水酶法油的抗氧化能力最强。综合来看,水酶法油的品质较好。(3)通过单因素和响应面法试验,得到超声波辅助法提取紫苏籽皮原花青素的最佳条件为:液料比15:1(m L/g),乙醇体积分数62%,超声时间30min,超声温度53℃,提取量为2.170mg/g。(4)通过静态吸附与解吸附试验,选择XDA-8树脂来纯化紫苏籽皮原花青素,经动态吸附与解吸附试验得出纯化的最优条件为:上样浓度4mg/mL,上样流速4BV/h,乙醇体积分数70%,洗脱流速2BV/h,洗脱剂的用量为13BV。纯化前的紫苏籽皮原花青素含量为5.25%,纯化后为12.10%,提高了130.48%。对纯化前后的紫苏籽皮原花青素、VC以及BHT四种样品的抗氧化能力进行测定与比较,发现纯化后的紫苏籽皮原花青素的抗氧化能力最强。
[Abstract]:Perilla is a traditional Chinese medicinal material and oil crop in China. Perilla seed has a high oil content and contains procyanidins in the skin, so the comprehensive utilization of perilla seed has a broad market prospect. In this paper, perilla seed oil was extracted from perilla seed by ultrasonic assisted water enzymatic method, and the properties of perilla seed oil were determined, and the extraction and purification of proanthocyanidins from perilla seed skin were studied. The antioxidant activity was determined. The main results are as follows: (1) on the basis of single-factor test, The optimum conditions for extracting perilla seed oil by ultrasonic assisted water enzymatic extraction were obtained by response surface method: ratio of complex enzymes (5. 5% of cellulase, 4.5% of neutral protease, 7.5% of Pectinase), liquid to material ratio of 6:1 (mL / g), ultrasonic time of 23 min, enzymatic hydrolysis temperature of 50 鈩,
