面向现场快速检测的高分辨Blu-ray成像技术的研究

【摘要】:蓝光光盘(Blu-ray Disc,BD)不仅可以作为全高清格式电影和大型电子游戏的存储媒介,并且凭借其优良的光学性能、纳米级的表面平整度,使其还可以作为化学分析和生物分子检测反应的基底材料,同时结合蓝光光驱精密的伺服控制系统和光学读取机制,蓝光光盘/光驱有望成为新一代的高通量、高灵敏度的现场快速检测(point-of-care testing,POCT)工具。本文以蓝光光驱读写原理和光盘存储技术为基础,通过提取、采集标准蓝光光驱内部光学头的光强信号,在Windows环境下研发了基于标准蓝光光盘/光驱的成像检测系统。通过测试打印高通量阵列BD光盘,验证了该系统的可行性、分辨率(至少可以达到100μm)以及检测通量(至少可以实现1728个待测样品同时检测),同时成功制备了黄曲霉毒素B1(Aflatoxin B1,AFB1)免疫优化反应BD光盘,并利用所研发的成像系统实现了AFB1抗体浓度的优化反应实验的定量检测。基于标准蓝光光盘/光驱的成像检测系统可以作为一个完整的检测平台应用于食品安全、水质监测和医疗诊断等重要领域。
[Abstract]:Blu-ray Disct BDB can be used not only as a storage medium for full-HD movies and large video games, but also for its excellent optical properties and nanometer surface smoothness. It can also be used as a substrate material for chemical analysis and biomolecular detection reactions. At the same time, combined with the precise servo control system and optical reading mechanism of Blu-ray optical drive, Blu-ray optical disc / optical drive is expected to become a new generation of high-throughput. High sensitivity on-the-spot rapid detection point-of-care testing POCTs. Based on the principle of reading and writing of Blu-ray optical drive and optical disk storage technology, this paper develops an imaging detection system based on standard Blu-ray optical disk / optical drive in Windows environment by extracting and collecting the light intensity signal of the optical head inside the standard Blu-ray optical drive. The feasibility of the system is verified by testing and printing high throughput array BD optical disk. At least 100 渭 m resolution (at least 100 渭 m) and detection flux (at least 1728 samples can be detected simultaneously, and aflatoxin B1(Aflatoxin B1 AFB1) immuno-optimization reaction optical disk (BD) was successfully prepared. The optimized reaction test of AFB1 antibody concentration was carried out by using the imaging system developed in this paper. The imaging detection system based on standard Blu-ray disc / optical drive can be used as a complete detection platform in important fields such as food safety, water quality monitoring and medical diagnosis.
