【摘要】:小肠结肠炎耶尔森菌是一类在自然界广泛分布的人兽共患病原菌,目前研究表明猪是其主要宿主,也是造成人类感染的主要来源。为了研究中国生猪中小肠结肠炎耶尔森菌的分布特征,于2009年3月至2011年11月期间选取位于中国中部的河南省;西南部的四川省、云南省;华北的北京市、内蒙古自治区、天津市;东北的黑龙江省;西北的宁夏回族自治区、青海省和华东的江苏省、江西省共11省区的大型屠宰场,养殖场及散养农户作为采样地点共采集8773份标本,其中屠宰场宰杀生猪咽拭子标本4495份,回盲部肠内容物标本1239份以及当地养殖场或农户散养猪粪标本3039份。细菌分离结果表明,小肠结肠炎耶尔森菌的平均带菌率从高到低依次为猪咽喉部19.53%,猪回盲部肠内容物7.51%和猪粪便5.30%。血清分型结果显示850株致病性菌株中除3株为2/0:9生物血清型,3株为4/0:3型外其他均为3/0:3型,可见0:3血清型已经取代0:9型成为我国致病性小肠结肠炎耶尔森菌的主要型别。从北京、天津、江西、江苏、宁夏回族自治区、内蒙古自治区、四川省区猪咽拭子中分离到的小肠结肠炎耶尔森菌均以致病性的3/0:3型为主(85.17%-100%),相反在东北部的黑龙江省猪咽拭子中分离到的菌株仅有3.48%(4/115)为3/0:3型,96.52%(111/115)为非致病性的生物1A型多种血清型混杂的菌株。对所有致病性菌株进行脉冲场凝胶电泳分析,结果表明847株0:3型致病性小肠结肠炎耶尔森菌菌共分为49个带型,其中以K6GN11C30021和K6GN11C30012带型占多数,分别为53.13%(450/847)和21.37%(181/847),且各个省市均有其独特的PFGE带型;同时通过聚类分析发现四川、江苏、天津、河南等地腹泻病人分离的致病性菌株与当地猪中分离株的主要带型一致。病人中小肠结肠炎耶尔森菌的PFGE带型组成与宿主动物猪、犬相似,而与其他家畜家禽则不大相同。 本研究结果表明中国多地区生猪中小肠结肠炎耶尔森菌的带菌率均较高,分离菌株的生物血清型分布具有明显地域性特征,并且通过分子生物学手段充分证实了猪的携带与病人感染的小肠结肠炎耶尔森菌密切相关。中国生猪养殖场和屠宰场分布广泛,人群食用猪肉普遍,应警惕致病性小肠结肠炎耶尔森菌通过猪感染人和其它动物以及环境的危险。
[Abstract]:Yersinia enterocolitica is a kind of common zoonotic bacteria which are widely distributed in nature. At present, studies have shown that pigs are the main hosts and the main source of human infection. In order to study distribution characteristics of bacteria in the small intestine of the pig China Yersinia colitis, during the period from March 2009 to November 2011 were located in central Henan province China; southwestern Sichuan Province, Yunnan Province, Beijing City, North China; the Inner Mongolia Autonomous Region, Tianjin; Northeast Heilongjiang province; northwest of the Ningxia Hui Autonomous Region, Qinghai province and East China's Jiangsu Province, Jiangxi province a total of 11 provinces of the large slaughterhouses, farms and backyard farmers as the sampling sites were collected 8773 specimens, including the slaughter hog slaughter 4495 throat swab specimens, ileocecus intestinal contents of 1239 samples and local farms or farmers backyard pig manure samples were 3039. The results showed that the bacteria isolation, average carrier yersiniaenterocolitica rate from high to low was pig throat 19.53% pigs, ileocecus intestinal contents of 7.51% and 5.30% pig manure. Serotyping results showed that 850 strains of pathogenic strains in 3 strains of 2/0:9 serotype 3 strains for biological, 4/0:3 and others are of type 3/0:3, visible 0:3 serotype has replaced 0:9 in China has become the main type of pathogenic Yersinia enterocolitica. Enterocolitis Jerson bacteria isolated in pharyngeal swabs from Beijing, Tianjin, Jiangxi, Jiangsu, the Ningxia Hui Autonomous Region, the Inner Mongolia Autonomous Region, Sichuan provinces that are pig disease type 3/0:3 (85.17%-100%), in contrast to the pig in the northeast of Heilongjiang province from pharyngeal swab in strains to only 3.48% (4/115) 3/0:3, 96.52% (111/115) for hybrid non pathogenic biotype 1A multiple serotype strain. By pulsed field gel electrophoresis analysis of all pathogenic strains, the results showed that 847 strains of type 0:3 pathogenic fungus enterocolitis Jerson is divided into 49 types, among which the K6GN11C30021 and K6GN11C30012 band majority, were 53.13% (450/847) and 21.37% (181/847), and each province has its unique PFGE band at the same time; through cluster analysis found that the main type strain of Sichuan, Jiangsu, Tianjin, Henan and other places separated from the diarrhea patients of pathogenic strains and local pigs. In patients with intestinal bacteria Yersinia colitis with PFGE type composition and host animal swine, dogs and other animals are similar, and are not the same.
