【摘要】:目的建立二乙基亚硝胺(DEN)诱导的小鼠肝癌模型并使用两种不同配方饲料:SPF级小鼠大鼠全价配合饲料和AIN93-G配方合成饲料分别喂养,探讨不同饲料喂养对构建DEN诱导小鼠肝癌模型的影响。方法14日龄雄性C57BL/6小鼠腹腔一次性注射DEN溶液,给药量25 mg/kg体重,建立小鼠肝癌模型。小鼠离乳后分为两组,一组以常规SPF级大小鼠全价配合饲料喂养,另一组以AIN-93G配方合成饲料喂养。模型小鼠生长至9月龄时处死,取肝脏组织称重,观察并记录肝癌生长情况。结果全价饲料组小鼠肝脏全部产生肿瘤,AIN-93G组小鼠体重和肝脏重量明显低于全价饲料组小鼠,肝癌发生率、肿瘤结节数量和大小也明显低于全价饲料组小鼠。结论全价饲料喂养的小鼠DEN诱导肝癌模型造模成功,而AIN-93G饲料喂养的小鼠诱导肝癌的发生发展受到抑制,同时小鼠体重明显下降,提示了饮食因素在动物疾病模型建立中起到的关键作用。
[Abstract]:Objective to establish two diethylnitrosamine (DEN) induced mouse HCC model and using two different feed formulations: SPF mice and rats the full price of feed and feed were fed formula AIN93-G synthesis, the effects of different diets on the construction of DEN induced mouse model of liver cancer. Methods 14 day old male C57BL/6 mice were intraperitoneally injection of DEN solution, the dosage of 25 mg/kg body weight, the establishment of mouse model. Mice after weaning were divided into two groups, one group with normal SPF level big mouse feed full price feed, another group with AIN-93G formula and synthetic feed. Death growth to September the old model of small rat liver tissue, weighing, observation and record the growth of hepatocellular carcinoma. Results the full price of feed, all mice liver tumors, body weight and liver weight in AIN-93G group was significantly lower than that of the full price of feed group, the incidence of hepatocellular carcinoma, tumor number and size also Ming The full price of feed group were significantly lower than DEN mice. Conclusion the full price of feed induced liver cancer model rats, while AIN-93G fed mice induced the development of HCC is inhibited, and body weight of mice decreased significantly, suggesting that dietary factors plays a key role in animal disease model establishment.
