补肾温阳祛风散寒法对肾阳虚型围绝经期模型大鼠ER、VEGF、HSP70作用的研究

【摘要】:目的:探讨补肾温阳祛风散寒法对肾阳虚型围绝经期模型大鼠ER、VEGF、HSP70的影响及作用机制。方法:选用自然老化的围绝经期雌性SD大鼠,肌注氢化可的松构建肾阳虚型围绝经期大鼠模型,将模型大鼠随机分为更年汤组、教材方组、替勃龙组、生理盐水组;另选青年组、老年组SD雌性大鼠作为对照组。各组给予相应药物治疗15 d,于第16天处死大鼠,分离摘取双侧子宫,称重了解子宫湿重及子宫指数,采用病理切片观察大鼠子宫形态学变化,免疫组化及原位杂交检测方法观察大鼠子宫雌激素受体(ER)、血管内皮生长因子(VEGF)、热休克蛋白70(HSP70)及信使mRNA(ERmRNA、VEGFmRNA、HSP70mRNA)的阳性表达情况。结果:1更年汤组、教材方组、替勃龙组能明显增加子宫湿重、子宫指数,有统计学意义(P0.05),更年汤组与替勃龙组、教材方组比较差异有统计学意义(P0.05);2更年汤组、教材方组、替勃龙组能抑制子宫萎缩,有显著性差异(P0.01),更年汤组与替勃龙组比较无统计学意义(P0.05);3更年汤组、教材方组、替勃龙组能增加ER、ERmRNA、VEGF、VEGFmRNA阳性表达,有统计学意义(P0.05),但教材方组的上调低于更年汤组及替勃龙组(P0.05),替勃龙组与更年汤组之间比较无显著差异(P0.05);4更年汤组、替勃龙组能增加HSP70及HSP70mRNA阳性表达,有统计学意义(P0.05),更年汤组与替勃龙组比较无显著性差异(P0.05)。结论:自拟更年汤可能通过上调ER、VEGF、HSP70蛋白及基因在子宫中的阳性表达而发挥雌激素样作用,从而达到改善病理状态下的子宫的形态结构及功能的目的。
[Abstract]:Objective: To investigate the kidney yang Qufeng method on kidney yang deficiency perimenopausal rat model of ER, VEGF, HSP70 and its mechanism. Methods: the natural aging of the perimenopausal female SD rats, injection of hydrocortisone in construction of kidney yang deficiency model peri climacteric rat model, the rats were randomly divided into Gengnian Decoction group, teaching group, tibolone group, saline group; another youth group and elderly group SD female rats as the control group. Each group was given corresponding drug treatment of 15 d on the sixteenth day, the rats were sacrificed and the separation from bilateral uterine, weighing about uterine wet weight and index of uterus, the pathological observation the morphological changes of uterus in rats, immunohistochemistry and in situ hybridization method to observe the rat uterine estrogen receptor (ER), vascular endothelial growth factor (VEGF), heat shock protein 70 (HSP70) and mRNA (ERmRNA, VEGFmRNA, Messenger HSP70mRNA) positive expression.
