人胚胎三叉神经节细胞形态发育研究

【摘要】:目的:观察人胚胎三叉神经节细胞形态结构及随胎龄增长变化发育规律。方法:取不同胎龄段人胚胎灌注固定后,外科显微镜下取双侧三叉神经节,常规HE染色,光镜观察;细胞计数;图像分析仪测量三叉神经节细胞直径、面积和周长;电镜下观察细胞内超微结构;比较不同胎龄时期细胞器变化情况;取1例标本在三叉神经根处植入DiI染色晶体,37℃恒温箱放置3个月,等待DiI染色晶体扩散,根据神经走向切片,荧光显微镜下观察神经元的形态;另取1例做冰冻切片神经元胞浆Nissl染色。结果:随着胎龄增长三叉神经节细胞数目无显著性变化;各胎龄组人胚胎三叉神经节神经细胞直径,19周至25周基本无变化,25周至32周发育过程中,平均直径增加最明显,面积和周长明显增大,与直径变化规律一致。电镜下,18-20周胎儿三叉神经节神经细胞排列紧密,胞质少,可见到数量不多的线粒体,且其内几乎未见嵴,其它细胞器较少。25周时,线粒体嵴变长,粗面内质网现雏形,有纵形小管出现;27周时可观察到成熟的高尔基复合体,32周时,线粒体、粗面内质网等细胞器结构发育趋于成熟;于33周电镜下可见溶酶体;36周时细胞内各种细胞器结构和功能基本完善;DiI追踪可清晰标记神经节细胞。结论:人胚胎三叉神经节细胞形态发育32周(8个月)时达成熟状态;三叉神经节细胞发育过程中随胎龄增加,其结构和功能逐步完善,32-36周(8-9月)是细胞的分化发育重要时期。
[Abstract]:Objective: to observe the morphological structure of trigeminal ganglion cells in human embryos and their developmental regularity with the increase of gestational age. Methods: the bilateral trigeminal ganglion was harvested under surgical microscope after different gestational age segments of human embryos were infused and fixed. Routine HE staining and light microscopic observation; Cell count; The diameter, area and perimeter of trigeminal ganglion cells were measured by image analyzer.
