大鼠骨髓间充质干细胞联合α-干扰素对大鼠肝损伤的治疗作用

【摘要】:目的:1建立大鼠肝损伤模型。 2在大鼠肝损伤模型的基础上初步探讨大鼠骨髓间充质干细胞联合α-干扰素对肝损伤的治疗作用。 方法:1用四氯化碳油溶液(50%体积分数)按2ml/kg对SD大鼠行腹腔注射,每周两次,共6周,建立大鼠肝损伤模型。于6周后随机挑选6只,处死大鼠,取肝组织行HE染色评价肝损伤程度。 2将肝损伤模型大鼠随机分为空白对照组(K)、大鼠骨髓间充质干细胞治疗组(S)、干扰素组(I)、大鼠骨髓间充质干细胞+干扰素治疗组(C)。分别于治疗后3天,6天处死大鼠,采血检测谷草转氨酶(AST)、谷丙转氨酶(ALT)、总胆红素(TBIL)、HA透明质酸酶、LN层粘连蛋白、PCⅢ型前胶原、Ⅳ型胶原,同时取肝组织行HE染色评价肝损伤严重程度。 结果:1大鼠肝损伤模型60只,死亡6只,存活54只死亡率为10%,存活的54只均出现腹围增加,随机选取6只取肝脏HE染色均提示有不同程度肝损伤,成功率为90%,符合本实验要求。 2大鼠慢性肝损伤后,AST指标显著高于ALT指标;药物治疗后,除了ALT浓度的干细胞治疗组和干扰素治疗组无差异外,AST、ALP、TBDL、LN,Ⅳ型胶原的组间对比均有显著性差异,联合治疗组浓度低于其余三组,且具有统计学差异。ALT,HA和Ⅲ型前胶原的浓度,联合治疗组浓度低于其余三组,且具有统计学差异,空白对照组和干细胞组相比有统计学差异,干细胞治疗组浓度低于空白对照组,干细胞治疗组浓度低于干扰素组,有统计学差异,空白对照组和干扰素组相比无统计学差异。 结论:1四氯化碳油溶液(50%体积分数)按2ml/kg对大鼠行腹腔注射,每周两次,共6周,建立肝损伤模型稳定可靠,为较理想的研究肝损伤模型。2α-干扰素和大鼠骨髓间充质干细胞对四氯化碳诱导的大鼠肝损伤有一定的治疗作用,干细胞治疗效果优于干扰素。3大鼠骨髓间充质干细胞联合α-干扰素对四氯化碳诱导的肝损伤的治疗有协同效果,联合治疗效果优于单药治疗。
[Abstract]:Objective to establish a rat model of liver injury by 1: 1. 2 on the basis of rat liver injury model, the therapeutic effect of bone marrow mesenchymal stem cells combined with interferon on liver injury was studied. Methods Sprague-Dawley rats were injected intraperitoneally with 50% volume fraction of carbon tetrachloride oil solution twice a week for 6 weeks. Six rats were randomly selected after 6 weeks. Rats were killed and liver tissues were taken to evaluate the degree of liver injury by HE staining. 2Rats with liver injury were randomly divided into control group (control group), rat bone marrow mesenchymal stem cell (BMSCs) treatment group and interferon group (group I). The rat bone marrow mesenchymal stem cell interferon treatment group was killed on 3 and 6 days after treatment, and blood samples were collected to detect aspartate and alanine aminotransferase (alt).
