去卵巢结合蔗糖铁诱导家兔铁蓄积状态骨质疏松模型的建立与评价

【摘要】:目的通过去卵巢结合蔗糖铁诱导家兔铁蓄积状态骨质疏松模型,以期为有关铁蓄积状态骨质疏松的实验研究提供动物模型基础。方法将30只5月龄的新西兰雌性家兔随机分为对照组、去势组和铁蓄积组;每组分别于术前1天检测血清1型前胶原氨基末端肽(P1NP),1型胶原羟基末端肽(CTX)及血清铁蛋白水平,并行腰椎骨密度测量;去势组和铁蓄积组手术切除双侧卵巢,对照组仅切开卵巢周围部分脂肪;1周后铁蓄积组耳缘静脉注射蔗糖铁,剂量为15 mg/kg,每周1次,共12周;对照组和去势组以同样方式和频次给予等量生理盐水注射;所有组分别于术后4、8、12、16、20、24周检测血清P1NP,CTX及血清铁蛋白水平,并行腰椎骨密度测量。结果 (1)所有家兔注射蔗糖铁后均未出现昏迷、抽搐、精神萎靡、呼吸急促等不良反应或致死的情况;(2)铁蓄积组血清铁蛋白水平在造模后8、12、16、20及24周均明显高于对照组(P0.05);(3)铁蓄积组与去势组血清P1NP,CTX水平在造模后12周、16周、20周及24周均明显高于对照组(P0.05);铁蓄积组血清P1NP,CTX水平在造模后20周及24周均明显高于去势组(P0.05);(4)铁蓄积组与去势组腰椎骨密度丢失百分率均在造模后第24周超过25%(33.72%、25.92%)。结论去卵巢结合蔗糖铁可成功建立家兔铁蓄积状态骨质疏松模型。
[Abstract]:Objective to induce the model of iron accumulation in rabbits by ovariectomized with sucrose iron. Methods Thirty 5-month-old New Zealand female rabbits were randomly divided into control group, castrated group and iron accumulation group. Serum procollagen type 1 amino terminal peptide (P1NPN) and serum ferritin (CTX) were measured 1 day before operation in each group. The bone mineral density of lumbar vertebrae was measured. The ovariectomized group and the iron accumulation group had bilateral ovariectomies, while the control group only cut some fat around the ovary. One week later, sucrose iron was injected intravenously into the ear margin of the iron accumulation group at a dose of 15 mg / kg once a week for 12 weeks. The control group and ovariectomized group were injected with the same amount of saline in the same way and frequency. The levels of serum P1NPCTX and serum ferritin were measured in all groups at 24 weeks after operation. Results all rabbits had no adverse reaction or death after injection of iron sucrose, such as coma, convulsion, mental retardation, shortness of breath, etc. (2) the level of serum ferritin in the iron accumulation group was significantly higher than that in the control group at 20 and 24 weeks after the establishment of the model. (3) the levels of serum P1NPN CTX in the iron accumulation group and castration group were significantly higher than those in the control group at the 12th week, the 16th week, the 20th week and the 24th week, respectively. The level of serum P1NPN CTX in the iron accumulation group was significantly higher than that in the castrated group at 20 and 24 weeks after the establishment of the model. 4) the percentage of bone mineral density loss of lumbar vertebrae in the iron accumulation group and castration group was more than 33.72% at the 24th week after the establishment of the model. Conclusion Ovariectomized with sucrose iron can successfully establish the model of iron accumulation in rabbits.
