Lnc-CLRN3与肠道天然免疫的相关性及其功能研究

【摘要】:目的研究一个全新的长链非编码RNA Lnc-CLRN3在肠道天然免疫中的作用。方法应用脂多糖(lipopolysaccharide,LPS)刺激人结肠癌细胞SW480,然后收集细胞提取RNA并通过RT-qPCR筛选表达变化的长链非编码RNA,进一步通过基因过表达、敲除和RNA免疫共沉淀等技术手段对该基因的功能进行深入分析。结果筛选出1个在SW480细胞中高表达且LPS刺激基因表达上调的长链非编码RNA Lnc-CLRN3,该基因主要分布于细胞核内,其转录受到MAPK通路的调控,进一步研究表明该基因通过结合SFPQ蛋白进而影响下游IL12P40等免疫因子的表达。结论长链非编码RNA Lnc-CLRN3参与了肠道细胞的天然免疫调控。
[Abstract]:Objective to study the role of a novel long chain noncoding RNA Lnc-CLRN3 in intestinal innate immunity. Methods Lipopolysaccharide lipopolysaccharide (LPS) was used to stimulate human colon cancer cell line SW480, and then the cells were collected to extract RNA and to screen the long chain noncoding for the change of expression by RT-qPCR. RNAs, further overexpression of genes, The function of the gene was analyzed by knockout and RNA immunoprecipitation. Results A long chain non-coding RNA Lnc-CLRN3, which was highly expressed in SW480 cells and up-regulated by LPS stimulating gene, was selected, and the gene was mainly distributed in the nucleus. Its transcription is regulated by MAPK pathway. Further studies indicate that the gene affects the expression of downstream IL12P40 by binding SFPQ protein. Conclusion long chain non-coding RNA Lnc-CLRN3 is involved in the innate immune regulation of intestinal cells.
