角叉菜胶诱导慢性非细菌性前列腺炎动物模型的制备与评价

【摘要】：目的建立化学性慢性非细菌性前列腺炎(chronic non-bacterial prostatitis,CNP)动物模型,为CNP的发病机制及药物研究提供可靠的模型动物及评价方法。方法将SD雄性大鼠随机分为对照组和模型A、B、C组,模型A、B、C组分别用1%的无菌角叉菜胶注射至前列腺左右腹侧叶中各20、50、100μL;对照组每只注入50μL灭菌的生理盐水溶液。7 d后处理,分别从解剖学角度、前列腺指数、白细胞检测、病理切片以及肿瘤坏死因子-α(TNF-α)、核因子-κB(NF-κB)、IκB激酶(IKKα)、磷酸化IKB-α(p-IKB-α)和环氧合酶-2(COX-2)蛋白表达的影响5个方面进行分析。结果与假手术组比较,模型A、B、C组的前列腺组织柔软程度下降,弹性减弱,与周围组织发生粘连;前列腺指数、白细胞总数均显著增加(P0.01),A、B、C组前列腺指数的增加率分别为21.1%、61.7%和72.7%;白细胞总数增加率分别为75.0%、103.6%和114.8%;病理结果显示A组前列腺间质肿胀较少,B、C组组间质明显肿胀,C组前列腺萎缩明显,部分腺体变性坏死;A、B、C组前列腺组织内NF-κB、IKKα、p-IKB-α、TNF-α和COX-2表达水平出现不同程度的增加,NF-κB炎症通道被激活。结论大鼠前列腺左右腹侧叶注射1%的角叉菜胶各20、50、100μL,均能够诱导不同程度前列腺炎症反应,但20μL剂量太小,炎症反应较弱,操作误差大;100μL炎症反应过强,有一定动物死亡;50μL剂量所造成的前列腺炎动物模型最为成功,并可明显激活NF-κB炎症信号通路。
[Abstract]:Objective to establish an animal model of chronic non-bacterial prostatitis in order to provide reliable animal model and evaluation method for the pathogenesis and drug study of CNP. Methods Sprague-Dawley male rats were randomly divided into two groups: control group and model group. The model group A was injected with 1%% aseptic carrageenin into the ventral lobe of the prostate, and the control group was treated with 50 渭 L sterilized saline solution for 7 days, and the prostate index and white blood cell were measured from the anatomic point of view. Pathological sections and the expression of TNF- 伪, NF- 魏 B, phosphorylated IKB- 伪, phosphorylated IKB- 伪 and cyclooxygenase-2 were analyzed. Results compared with sham-operated group, the tissue softness of prostate was decreased in the model group. Reduced elasticity, adhesion to surrounding tissue; prostate index, The increase rate of prostatic index in group C was 21.1% and 72.7%, respectively, and the increase rate of total leukocyte count was 75.0% and 114.8%, respectively. The pathological results showed that the interstitial tumefaction in group A was less than that in group B C, and the prostatic atrophy in group C was obvious. The expression levels of NF- 魏 BK 伪 -IKK 伪 -IKB- 伪 -IKB- 伪 and TNF- 伪 and COX-2 were increased to varying degrees in the prostatic tissues of partial adenodegenerative necrotic group. Conclusion the inflammatory channels of NF- 魏 B are activated by injecting 1% carrageenin into the left and right ventral lobes of the prostate in rats, all of which can be induced to be different. Degree of prostatic inflammation, However, the dose of 20 渭 L was too small, the inflammatory response was weak, and the inflammatory response of 100 渭 L was too strong. The animal model of prostatitis caused by a certain dose of 50 渭 L of animal death was the most successful, and the NF- 魏 B inflammatory signal pathway could be activated obviously.
