丁基苯酞对痴呆模型大鼠认知能力及线粒体功能的作用

【摘要】:目的观察丁基苯酞对Aβ25-35致痴呆大鼠学习记忆的改善情况,并初步探索其作用机制。方法实验设立正常组、模型组和丁基苯酞组(10、30、100mg/kg),模型组和丁基苯酞组均采用侧脑室注射Aβ25-35,干预组造模后给予丁基苯酞灌胃(灌胃剂量为2.5mL/kg)。利用水迷宫实验观察大鼠的学习记忆能力,采用ATP检测观察脑组织内线粒体功能的变化,并用试剂盒检测线粒体相关酶活性的变化。结果水迷宫实验结果显示,模型组大鼠学习记忆能力与正常组比较差异有统计学意义(P0.05),丁基苯酞10mg/kg组、30mg/kg组大鼠学习记忆障碍均有明显改善(P0.05),100mg/kg组改变不明显;线粒体相关检测发现,与正常组相比,模型组ATP水平明显下降(P0.05),线粒体呼吸链复合物Ⅳ(P0.05)、丙酮酸脱氢酶复合体、α-酮戊二酸脱氢酶活性均明显降低(P0.05)。与模型组相比,丁基苯酞能明显提高线粒体相关酶的活性从而改善线粒体功能。结论丁基苯酞可以通过改善线粒体功能,提高Aβ25-35致痴呆大鼠的学习记忆能力。
[Abstract]:Objective to observe the effect of Ding Ji phthalide on learning and memory of dementia rats induced by A 尾 25-35, and to explore its mechanism. Model group and Ding Ji phthalide group (1030g 路kg ~ (-1) 路kg ~ (-1)), model group and Ding Ji phthalide group were treated with intracerebroventricular injection of A 尾 _ (25-35), then the intervention group was treated by intragastric instillation of Ding Ji phthalide at a dose of 2.5 mL / kg 路kg ~ (-1). The learning and memory abilities of rats were observed by water maze test. The changes of mitochondrial function in brain tissue were observed by ATP, and the activity of mitochondrial related enzymes was detected by kit.
