姜黄素壳聚糖微球制备及对2型糖尿病大鼠模型糖脂代谢的影响

【摘要】:目的制备姜黄素壳聚糖微球并研究其对2型糖尿病模型大鼠糖脂代谢的影响。方法用乳化分散-戊二醛交联法制备姜黄素壳聚糖微球。高脂饲料连续喂养SD大鼠4周后,腹腔注射链脲佐菌素(25 mg·kg~(-1)),建立2型糖尿病大鼠模型。将大鼠随机分为正常组、模型组和3个剂量实验组(姜黄素壳聚糖微球,50,150,250 mg·kg~(-1))共5个组。于给药前后,测定空腹血糖、空腹胰岛素、餐后2 h血糖等指标,以及测定血清中总胆固醇(TC)、三酰甘油(TG)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL)含量;并用荧光定量PCR技术检测血清中7个microRNA分子的表达量。结果制备的微球粒径均匀,载药量合适。给药后,与正常组比较,模型组、实验组大鼠的FBG、2PBG含量明显升高;血清中TC、TG、LDL水平均明显增高,HDL水平下降;microRNA的表达量也明显上调(均P0.05)。高剂量实验组组的疗效更佳。结论姜黄素壳聚糖微球的制备简单方便,重复性好,其可改善2型糖尿病大鼠中的糖脂代谢紊乱的状况。
[Abstract]:Objective to prepare curcumin chitosan microspheres and study the effect of curcumin chitosan microspheres on glycolipid metabolism in type 2 diabetic rats. Methods curcumin chitosan microspheres were prepared by emulsification dispersion and glutaraldehyde crosslinking. The rat model of type 2 diabetes mellitus was established by intraperitoneal injection of streptozotocin 25 mg 路kg ~ (-1). The rats were randomly divided into normal group, model group and three experimental groups (curcumin chitosan microspheres 5050,250mg 路kg ~ (-1)). Fasting blood glucose was measured before and after administration of streptozotocin. Fasting insulin, 2 h postprandial blood glucose, and serum total cholesterol TCN, triglyceride TGN, high density lipoprotein (HDL), low density lipoprotein (LDL) were measured. Fluorescence quantitative PCR technique was used to detect the expression of seven microRNA molecules in serum. Results the microspheres prepared were of uniform size and suitable drug loading. Compared with the normal group, the FBG-2PBG content in the model group and the experimental group was significantly higher than that in the normal group. The level of TCU TGN LDL in serum was significantly increased and the level of HDL decreased. The expression of microRNA was also upregulated (P 0.05). Conclusion the preparation of curcumin chitosan microspheres is simple, convenient and reproducible. It can improve the disorder of glucose and lipid metabolism in type 2 diabetic rats.
