没食子提取物对IgA肾病模型大鼠的治疗作用

【摘要】:目的研究没食子提取物(TGE)对Ig A肾病(Ig AN)模型大鼠血清、尿液及肾组织Ig A表达的影响。方法 50只健康雄性Sprague-Dawley大鼠随机分为正常对照组、Ig AN模型组、TGE 75,150和300 mg·kg~(-1)治疗组,每组10只。除正常对照组外,其余各组采用牛血清白蛋白(BSA)+脂多糖(LPS)+四氯化碳(CCl_4)联合造模,持续12周。从第13周开始,ig给予TGE,正常对照组及Ig AN模型组给予等量生理盐水,持续4周。第12周末及第16周末用BCA法测定24 h尿蛋白;第16周末,酶联免疫吸附法(ELISA)测定血清及尿液Ig A水平,全自动生化分析仪检测血清肌酐(SCr)和尿素氮(BUN)水平,按牛津分型对肾组织病理进行评分,免疫荧光法测定肾组织Ig A免疫复合物沉积。结果与正常对照组相比,Ig AN造模大鼠在12周末24 h尿蛋白升高(P0.05);在16周末,Ig AN模型组大鼠24 h尿蛋白、血清和尿液Ig A水平、血清SCr和BUN水平、肾组织牛津病理评分和Ig A沉积均较正常对照组升高(P0.05);与Ig AN模型组相比,TGE各给药组大鼠24 h尿蛋白、血清和尿液Ig A水平以及血清SCr水平降低(P0.05),TGE 150和300 mg·kg~(-1)组血清BUN水平和肾组织Ig A含量下降(P0.05),仅TGE 300 mg·kg~(-1)组牛津病理评分中有明显减少。结论 TGE可减少Ig AN模型大鼠血清、尿液及肾组织Ig A水平,对Ig AN具有一定程度的治疗作用。
[Abstract]:Objective to study the effect of TGE on the expression of IgA in serum, urine and renal tissue of rats with IgA nephropathy. Methods Fifty healthy male Sprague-Dawley rats were randomly divided into two groups: normal control group (Ig an model group) treated with TGE 75,150 mg 路kg ~ (-1) and control group (300 mg 路kg ~ (-1)). 10 rats in each group were treated with BSA (BSA) and CCl4 (CCl4) in each group for 12 weeks. TGEI was given orally at the 13th week, and the normal control group and the IgAN model group were given the same amount of normal saline, and the control group and the IgAN model group were given the same amount of normal saline, and the control group and the IgAN model group were given the same amount of normal saline. Serum and urine levels of IgA were determined by Elisa at the end of week 16, serum creatinine (creatinine) and bun (bun) were measured by automatic biochemical analyzer. According to the Oxford classification, the renal tissue pathology was graded, Results compared with the normal control group, the urinary protein of IgAN model rats increased P0.05A at the end of 12 weeks and the levels of urinary protein, serum and urine IgA at the end of 16 weeks in the IgAN model group. The levels of serum SCr and BUN, Oxford pathological score and IgA deposition in renal tissue were higher than those in normal control group (P 0.05), and 24 h urine protein of rats in Ig an model group were higher than those in control group. Serum and urine IgA levels and serum SCr levels decreased in P0.05TGE150 and 300mg 路kg-1) groups. The levels of serum BUN and renal tissue IgA decreased P0.05a, only TGE 300mg 路kg-1) group showed a significant decrease in Oxford pathological score. Conclusion TGE can reduce the serum level of IgAN model rats. The level of IgA in urine and renal tissue has some therapeutic effect on IgAN.
