结构信息对遗传关联分析影响的模拟研究

【摘要】:在复杂疾病基因定位研究中,人们会获得许多与之相关的表现性状,我们称之为多维疾病性状。它们可能由相同的遗传变异导致,也可能受不同的遗传变异控制。虽然基于多维性状的关联分析方法较复杂,基于单个疾病性状的关联分析方法较为简单,但是在某些情况下,单个疾病性状的关联分析方法会降低检验的功效。已经有人通过模拟研究指出在什么情况下采取多维疾病性状的关联分析功效性要优于单个疾病性状的关联分析。但是以往进行多维性状关联分析时,,虽然考虑到了性状之间的关系,却并没有考虑过性状间的结构信息,即性状间的方向性对分析效果的影响。本文我们通过考虑致病位点与性状间的多种关系结构图来产生模拟数据,通过模拟研究来分析性状间的结构信息对遗传关联分析的影响。模拟结果显示,当基因与性状间关联性较弱时,考虑性状间的结构信息优势并不显著;基因与性状间关联性越强,其功效性优势越显著。
[Abstract]:In the study of complex disease gene mapping, people will get a lot of related characters, we call multidimensional traits. They may cause disease by the same genetic variation, genetic variation may also be affected by different control. Although the correlation analysis method of multidimensional complex traits based on correlation analysis method based on single disease traits relatively simple, but in some cases, relevance of single disease traits analysis method will reduce the effectiveness of test has been through simulation studies and points out that adopting correlative multi-dimensional disease traits in what circumstances the association analysis is better than that of single disease traits to analysis efficacy. But previous analysis of multidimensional association, while taking into account the relationship between the traits, but did not consider structural information between characters, effects between traits of direction effect analysis. In this paper we Considering the variety structure of the relationship between characters and pathogenic sites to generate simulated data, through simulation studies to analyze the impact of structural information of the traits of genetic association. The simulation results show that when the relationship between genes and traits is weak, considering the structural information advantage among traits is not significant; the correlation between the genes and traits strong, its efficacy advantage is more significant.
