佐剂提高过敏反应动物模型敏感性研究

【摘要】:目的:通过观察茵栀黄注射液和生脉注射液佐剂组与未联用佐剂组在过敏反应发生时的敏感指标的含量变化,研究此类佐剂提高BN大鼠过敏反应动物模型稳定性和灵敏度的能力。方法:将弗氏完全佐剂与双黄连注射液、生脉注射液联用,对BN大鼠进行致敏和激发,比较联用弗氏佐剂组与未加弗氏佐剂组动物在过敏反应发生率、过敏反应发生程度;激发前后血浆中组胺、类胰蛋白酶、β-氨基己糖苷酶、Ig E含量及增长率;被动皮肤过敏试验大鼠皮下是否出现蓝斑以及蓝斑面积大小。结果:(1)联用佐剂组过敏反应的发生率以及过敏反应的发生程度均高于未加佐剂组;(2)茵栀黄注射液组激发前后血浆中组胺、类胰蛋白酶、β-氨基己糖苷酶、Ig E的含量与生理盐水组比较无显著性差异;联用佐剂后血浆中Ig E、组胺以及β-氨基己糖苷酶的含量均升高,与生理盐水组相比具有显著性的差异,激发前后四种指标的变化率均高于茵栀黄注射液未加佐剂组;生脉注射液组激发前后血浆中四种指标的含量与生理盐水组比较无显著性差异;联用佐剂后血浆中Ig E、β-氨基己糖苷酶的含量均升高,与生理盐水组相比具有显著性的差异,激发前后四种指标的变化率均高于生脉注射液未加佐剂组;(3)联用佐剂组的被动皮肤过敏反应的阳性率均高于未加佐剂组。结论:弗氏完全佐剂可提高过敏反应动物模型的敏感性,有利于中药注射剂安全性中过敏反应的再评价。
[Abstract]:Objective: to observe the changes of sensitive indexes in the allergic reaction of Yinzhihuang injection, Shengmai injection adjuvant group and uncombined adjuvant group.To study the ability of this adjuvant to improve the stability and sensitivity of allergic reaction animal model in BN rats.Methods: BN rats were sensitized and stimulated by the combination of Freund's complete adjuvant and Shuanghuanglian injection and Shengmai injection. The incidence of allergic reaction and the degree of allergic reaction were compared between Freund's adjuvant group and non-Freund 's adjuvant group.Plasma histamine, trypsin, 尾 -aminohexosaminidase (尾 -glucosidase) IgE content and growth rate were detected before and after stimulation, and whether there were locus coeruleus and the size of locus coeruleus in passive skin allergy test rats.Results the incidence of allergic reaction and the degree of anaphylaxis in the adjuvant group were higher than those in the non-adjuvant group. The histamine in the plasma of the Yinzhihuang injection group was higher than that of the control group before and after stimulation.The change rate of the four indexes before and after stimulation was higher than that of Yinzhihuang injection without adjuvant group, and there was no significant difference between Shengmai injection group and physiological saline group before and after stimulation.The plasma levels of IgE, 尾-hexosaminidase increased after combined use of adjuvant, and there was a significant difference compared with the saline group.The change rates of the four indexes before and after stimulation were higher than those in the non-adjuvant group of Shengmai injection and the positive rate of the passive skin allergic reaction in the combined adjuvant group were higher than those in the non-adjuvant group.Conclusion: Freund's complete adjuvant can improve the sensitivity of allergic reaction animal model and be helpful to reevaluate the safety of traditional Chinese medicine injection.
