左归丸联合温和灸对骨质疏松症模型大鼠骨密度、骨保护素mRNA及核因子κB受体活化因子配体mRNA的影响

【摘要】:目的探讨左归丸联合温和灸治疗骨质疏松症的可能作用机制。方法 40只雌性SD大鼠随机分为空白组、模型组、左归丸组、左归丸+温和灸组,每组10只。模型组、左归丸组、左归丸+温和灸组采用手术完整摘除双侧卵巢法制造骨质疏松症模型。造模3个月后左归丸组予左归丸10 ml/(kg·d)灌胃,左归丸+温和灸组予左归丸10 ml/(kg·d)灌胃+长强穴艾灸治疗,空白组和模型组予生理盐水10 ml/(kg·d)灌胃。各组均干预6周。干预前后测定大鼠股骨近端及第2腰椎骨密度,干预后通过PCR法检测大鼠骨组织骨保护素(OPG)mRNA和核因子κB受体活化因子配体(RANKL)mRNA表达水平。结果与空白组比较,模型组股骨、腰椎骨密度,骨组织OPG mRNA表达及OPG/RANKL降低,RANKL mRNA表达上升(P0.05或P0.01);与模型组比较,左归丸组、左归丸+温和灸组大鼠股骨、腰椎骨密度,骨组织OPG mRNA表达及OPG/RANKL均升高,RANKL mRNA表达降低(P0.05或P0.01),且左归丸+温和灸组大鼠股骨、腰椎骨密度,骨组织OPG mRNA表达及OPG/RANKL均较左归丸组升高,RANKL mRNA表达较左归丸组降低(P0.05)。结论左归丸联合温和灸能明显升高骨质疏松症大鼠的骨密度,上调OPG mRNA表达、下调RANKL mRNA表达可能是其作用机制之一。
[Abstract]:Objective to explore the possible mechanism of Zuo Gui Pill combined with mild moxibustion in the treatment of osteoporosis. Methods Forty female Sprague-Dawley rats were randomly divided into blank group, model group, Zuo Gui Wan group and Zuo Gui Wan mild moxibustion group with 10 rats in each group. Model group, Zuo Gui Wan group and Zuo Gui Wan mild moxibustion group were treated with complete ovariectomy to make osteoporosis model. After 3 months, Zuo Gui Pill group was given Zuo Gui Pill (10 ml/(kg d), Zuo Gui Wan mild moxibustion Group (ZJW 10 ml/(kg) was treated with Changqiang Point moxibustion, and the blank group and model group were treated with normal saline for 10 ml/(kg d). All groups were treated for 6 weeks. Bone mineral density (BMD) of proximal femur and 2 lumbar vertebrae were measured before and after intervention. After intervention, the expression of osteoprotegerin (OPG) mRNA and nuclear factor- 魏 B receptor activator ligand (RANKLN) mRNA were detected by PCR method. Results compared with the blank group, the bone mineral density (BMD) of femur and lumbar vertebrae, the expression of OPG mRNA in bone tissue and the decrease of mRNA expression of RANKL in the model group were increased (P0.05 or P0.01), compared with the model group, the bone mineral density of femur and lumbar vertebrae in the model group, Zuo Gui Wan mild moxibustion group, and Zuo Gui Wan mild moxibustion group were increased. The expression of OPG mRNA and OPG/RANKL in bone tissue were increased. The expression of RANKL mRNA was decreased (P 0.05 or P 0.01). The bone mineral density (BMD), bone tissue OPG mRNA expression and OPG/RANKL expression in the mild moxibustion group were higher than those in the Zuo Gui Wan group. The expression of RANKL mRNA was lower than that of the left Gui Wan group (P 0.05). Conclusion Zuo Gui Pill combined with mild moxibustion can significantly increase bone mineral density (BMD), upregulate OPG mRNA expression and down-regulate RANKL mRNA expression in osteoporosis rats.
