蓝莓联合益生菌对非酒精性脂肪性肝病模型大鼠PNPLA3和SREBP-1c表达的影响

【摘要】:目的观察含patatin样磷脂酶域3(PNPLA3)和胆固醇调节元件结合蛋白(SREBP)1c在蓝莓联合益生菌对非酒精性脂肪性肝病(NAFLD)模型大鼠干预实验中的表达,并探讨其在NAFLD中的作用机制。方法清洁级SD大鼠36只分为正常组(NG)、模型组(MG)、自然恢复组(SRG)、蓝莓组(BG)、益生菌组(PG)和蓝莓联合益生菌组(BPG)。实验结束后,制作组织切片,观察血清学、组织学形态变化,检测PNPLA3和SREBP-1c基因及蛋白表达情况。多组间比较用单因素方差分析,方差齐时用SNK-q检验,方差不齐时用Tamhane's-T2检验。结果 6组间在肝脏指数、ALT、TC、TG、LDL和HDL上差异均有统计学意义(F值分别为384.908、188.554、75.523、94.667、95.235、132.586,P值均0.01)。6组间NAFLD活动度积分差异有统计学意义(F=71.896,P0.01),其中BPG组较MG、SRG、BG、PG组下降最为显著(P值均0.01)。6组肝组织表达PNPLA3和SREBP-1c阳性率差异均有统计学意义(F值分别为175.527、110.504,P值均0.01),其中BPG组较MG、SRG、BG、PG组下降最为显著(P值均0.01)。PNPLA3和SREBP-1c的mRNA和蛋白表达各组间差异有统计学意义(F值分别为375.822、410.379、288.940、116.054,P值均0.01)。其中BPG组的PNPLA3和SREBP-1C的mRNA和蛋白表达较MG、SRG、BG、PG组明显下降(P值均0.01)。结论蓝莓联合益生菌能有效改善NAFLD的病理组织结构,减轻肝细胞脂肪变性,其机制可能是蓝莓与益生菌联合可减轻炎症因子诱发的炎症效应,下调SREBP-1c的表达,从而减弱PNPLA3基因转录,增强胆固醇代谢,减少脂质沉积,是NAFLD的一个辅助治疗方案。
[Abstract]:Objective to observe the expression of patatin-like phospholipase domain 3 (PNPLA3) and cholesterol regulatory element binding protein (SREBP1c) in the intervention experiment of blueberry combined with probiotics in rats with nonalcoholic fatty liver disease (NAF LDD) and to explore the mechanism of its role in NAFLD. Methods Thirty-six SD rats of clean grade were divided into normal group, model group, natural recovery group, blueberry group, probiotics group (PGN) and blueberry combined probiotics group. At the end of the experiment, tissue sections were made, serological and histological changes were observed, and the expression of PNPLA3 and SREBP-1c genes and proteins were detected. Single factor analysis of variance (ANOVA) was used in multigroup comparison, SNK-q test was used when variance was homogeneous, and Tamhane's-T2 test was used when variance was uneven. Results there were significant differences in NAFLD activity scores between the six groups on the liver index (alt) and HDL (P = 384.908188.554, P = 75.5239.667.65.235132.586p, respectively). There was a significant difference in NAFLD activity between the six groups (F71.896a, P0.01P). The BPG group had the most significant decrease compared with the MGG group (P < 0.01). The difference of PNPLA3 and SREBP-1c positive rate was statistically significant (F = 175.527110.504 P = 0.01), in which the mRNA and protein expression of 0.01).PNPLA3 and SREBP-1c in BPG group were significantly lower than those in MGG BPG group (P = 375.822410.379, 288.940116.054, P < 0.01). The expression of mRNA and protein in PNPLA3 and SREBP-1C in BPG group was significantly lower than that in MGG group. Conclusion the combination of blueberry and probiotics can effectively improve the pathological structure of NAFLD and reduce the steatosis of hepatocytes. The mechanism may be that the combination of blueberry and probiotics can reduce the inflammatory effect induced by inflammatory factors and down-regulate the expression of SREBP-1c. It is an adjuvant therapy for NAFLD to reduce the transcription of PNPLA3 gene, enhance cholesterol metabolism and reduce lipid deposition.
