Y染色体6个STR基因座在中国朝鲜族男性和汉族男性人群中遗传多态性

【摘要】:目的:研究Y染色体6个STR基因座DYS19、DYS388、DYS390、DYS391、DYS392、DY8393的遗传多态性在中国朝鲜族男性和汉族男性人群中的分布特征,并为法医学个体识别、亲权鉴定、人类遗传学及其他相关学科提供参考数据。 方法:(1)收集200名无血缘关系的朝鲜族男性和200名汉族男性人群的抗凝血,用小量全血基因组DNA快速提取试剂盒提取DNA。(2)采用PCR扩增结合5%聚丙烯酰胺凝胶电泳技术和银染显带方法对200名朝鲜族男性和200名汉族男性的DYS19、DYS388、DYS390、DYS391、DYS392、DYS393基因座进行检测,并用Arlequin2000软件进行数据分析.(3)对6个基因座的等位基因分别进行测序,得到等位基因分型标准物,并根据国际法医遗传学会DNA委员会(International Society of Forensic Genetics ISFG.DNA Commission)推荐命名. 结果:基因座DYS19、DYS388、DYS390、DYS391、DYS392、DYS393,在朝鲜族男性人群中依次检测到6、4、5、5、6、5个等位基因,遗传多态性(gene diversityGD)依次为0.6722、0.4848、0.6674、0.2641、0.6704、0.6405,将这6个Y-STR基因座累计分析,一共检测到109种单倍型,累计GD值为0.9803;在汉族男性人群中依次检测到6、5、7、5、7、5个等位基因,遗传多态性(gene diversityGD)依次为0.7130、0.5219、0.6971、0.3509、0.7439、0.6564,将6个基因座累计分析,一共检测到133种单倍型,累计GD值为0.9914。 结论:从本文数据显示,中国朝鲜族男性和汉族男性人群6个Y-STR基因座,除DYS391遗传多态性比较低外,其他基因座遗传多态性分布较好,稳定性强,具有较高的应用价值,可为中国朝鲜族男性和汉族男性人群的法医学个人识别,亲权鉴定,遗传学及其他相关学科提供参考数据。
[Abstract]:Objective: to study the distribution of the genetic polymorphism of DYS19 / DYS388nDYS390DYS391and DYS392DY8393 in 6 STR loci of Y chromosome in Chinese male population of Korean and Han nationality, and to identify them as forensic individuals and to identify their paternity, and to investigate the relationship between DYS388and DYS390DYS391and DYS392DYS392DY8393. Human genetics and other related disciplines provide reference data. Methods the anticoagulant blood samples were collected from 200 unrelated Korean males and 200 Han males. The DYS19DYS388DYS391DYS391DYS392DYS392DYS391DYS391DYS392DYS393 locus of 200 Korean and 200 Han Chinese men were detected by PCR amplification, 5% polyacrylamide gel electrophoresis and silver staining. The alleles of 6 loci were sequenced with Arlequin2000 software, and the standard alleles were obtained and named according to the recommendations of the DNA Committee of the International Society of Forensic Genetics. Results: DYS19, DYS388, DYS390, DYS391DYS392and DYS393 were detected in the Korean male population, and 5 alleles were detected in turn, and the genetic polymorphism gene diversityGDwas 0.672222 0.48480.66774 (0.2641), 0.670441 (0.670441) and 0.6704405. the six Y-STR loci were analyzed and 109 haplotypes were detected. The accumulative GD value was 0.9803, and the genetic polymorphism gene diversityGDwas 0.7130 / 0.52190.6971 / 0.35090.439/ 0.65644.A total of 133 haplotypes were detected in the six loci of Han nationality, with a total of 133 haplotypes (GD = 0.9914). Conclusion: according to the data of this paper, there are 6 Y-STR loci in Chinese Korean and Han male populations, except for the low DYS391 genetic polymorphism, the other loci are more stable and have higher application value. It can provide reference data for forensic personal identification, paternity identification, genetics and other related subjects of Chinese Korean and Han male populations.
